Abstract The basic scheme of the taxonomy of cyanobacteria (cyanophyceae blue-green algae) widely... more Abstract The basic scheme of the taxonomy of cyanobacteria (cyanophyceae blue-green algae) widely known and accepted is the classical treatment by the phycologists of the past several decades. Identification was based on critical microscopic examination and illustrations along with the information on the habitat. The revision of the cyanophytes by Drouet and his coworkers incited much interest in the taxonomy of blue-green algae in India. Earlier studies brought out the distinction and plasticity of the organisms under culture condition, even to the level of questioning the identity of certain genera or species. The terminology of cyanobacteria for Cyanophyceae resulted in considerable disagreement among phycologists. Newer characteristics like the fatty acid composition, molecular percentage of G/C and fine structure of cellular constituents were suggested to delineate the taxa. The ultrastructure of photosynthetic lamellae of unicellular and filamentous forms was analyzed. Modern molecular techniques such as DNA sequencing, random amplified polymorphic DNA, DNA polymorphisms, genomic fingerprinting technique, RFLP of psbA genes and the applicability of STRR, RAPD, and REP-PCR fingerprinting for the strain level differentiation have been used by several scientists to distinguish strains at the species and genus level. DNA amplification fingerprinting has been very useful for distinguishing closely related genotypes. RFLP markers are used for characterizing genetic diversity. Other tools have been 16S rRNA sequences, amplified 16S–23S rDNA spacer (ITS-1). Recent developments are the polyphasic analysis of the cyanobacteria from extreme habitats. The classification of the organisms under the presently well-known treatment of the group as Cyanoprokaryota is appended.

The ability of nodal shoot segments regenerated in vitro to form plantlets under culture conditio... more The ability of nodal shoot segments regenerated in vitro to form plantlets under culture conditions has been investigated in Glycine clandestina, a wild relative of cultivated soybean (Glycine max). The nodal shoot segments (NSSs) were excised from in vitro regenerated shoots which differentiated from the cultured cotyledons on B5 medium supplemented with IBA (0.05 mg/l) and BAP (3.0 mg/l). Plant regeneration from NSSs occurred through direct multiple shoot as well as via callus formation on B5 medium with different levels of IBA (0.005 to 0.05 mg/l) and BAP (1 to 10 mg/l). Clumps of the regenerated shoot segments were further subcultured on B5 medium with IBA (0.005 mg/l) and BAP (1.0 mg/l) for continuous shoot proliferation. Plants can be obtained from shoot buds when transferred to low levels of IBA and BAP. Cyanobacteria are ancient photosynthetic organisms inhabitant of various habitats including arid ... more Cyanobacteria are ancient photosynthetic organisms inhabitant of various habitats including arid and semi- arid water bodies. These are effective producers of various biologically active compounds that have been widely used in food, medicine, cosmetics and pharmaceutical industry. However, non-heterocystous cyanobacteria of arid and semi- arid water bodies are less explored for biotechnological applications. The aim of the present study was to investigate non-heterocystous filamentous cyanobacteria of freshwater bodies of semi-arid region for their pigment profile, phenolic contents and antioxidant properties. Our results revealed that among 8 cyanobacterial strains, D. tharense AT2015/7 and S. subsalsa AT2016/1 possessed higher amount of valuable pigments, phenolic contents and also showed good antioxidant properties. S. subsalsa AT2016/1 was the most promising cyanobacterial taxa for the production of phycocyanin when compared to so far studied non-heterocystous cyanobacteria inhabit aquatic habitats. In the LC-HRMS/MS analysis, 11 phenolic compounds were tentatively identified in D. tharense AT2015/7 and nine phenolic compounds detected in S. subsalsa AT2016/1. The present study revealed that cyanobacteria thriving in the semi-arid region could be promising candidates for the production of valuable pigments, phenolic compounds and could be a potential source of antioxidants.

Salt-loving microorganisms can flourish in saline environments which inhibit the growth of other microbial life, and they possess the potential to produce stable and novel biomolecules for the use in biotechnological applications, including anticancer compounds. Sambhar Lake is the largest inland soda lake in India and is an appropriate habitat for halophilic bacterial and archaeal strains in terms of diversity and potential production of bioactive compounds. In the present study, a moderately halo-alkaliphilic bacterial strain C12A1 was isolated from Sambhar Lake, located in Rajasthan, India. C12A1 was gram-positive, motile, rod-shaped, formed oval endospores, produced carotenoids, and exhibited optimal growth at 37 °C in 10–15% NaCl (pH 8). Cyanobacteria are one of the promising organisms to produce a number of secondary metabolites that are capable of reducing silver ions to small-sized silver nanoparticles. In the present study, we employed an aqueous extract of the cyanobacterium Haloleptolyngbya alcalis KR2005/106 isolated from a soda lake for biosynthesis of silver nanoparticles (AgNPs). The extract acted as a reducing agent for AgNPs synthesis and resulted formation of nanoparticles < 50 nm in size. In this study, synthesis of AgNPs obtained only in the sample exposed to photosynthetically active radiation (PAR) while the synthesis of AgNPs was not observed in the samples kept in dark. The biogenic fabrication of AgNPs was carried out by optimizing several governing parameters such as concentration of the silver nitrate solution, pH, temperature, and amount of biomass. Results obtained through different analytical techniques revealed that cyanobacterial taxon H. alcalis isolated from saline-alkaline habitat is a potential candidate for biosynthesis of optimum-sized spherical AgNPs. Surface plasmon resonance (SPR) property of AgNPs was exploited for aqueous ammonia sensing and revealed that AgNPs synthesized using aqueous extract of cyanobacterium H. alcalis could be employed for colorimetric detection of dissolved ammonia for monitoring quality of water.

Microphytes (Cyanobacteria and algae) are amongst the most promising sustainable resources for hi... more Microphytes (Cyanobacteria and algae) are amongst the most promising sustainable resources for high-quality compounds, but high expenditures for culturing, harvesting and subsequent processing complicate their exploitation. An economic way to reduce costs of algal mass cultures is to install open-pond culture systems in tropical regions, which guarantee high solar irradiance throughout the year, but this approach is still waiting to be introduced in East Africa. One essential but often neglected requirement to carry out successful microphytean mass cultures is strain selection. We provide a selection of high-yielding microphytes with high potential for biotechnology use collected in the soda lakes and adjacent systems such as salt evaporation ponds or sewage oxidation ponds. Microphytes were detected as field clones or cultured strains and identified via microscopy and molecular methods; SSU rRNA and/or ITS sequences were entered in the NCBI database. Besides the well-known and already commercially exploited cyanobacterium Arthrospira, promising relatives of the famous “green balls” Chlorella, members of the green algal genera Selenastrum, Picocystis and Mychonastes and the eustigmatophytes Nannochloropsis and Microchloropsis are presented. All of them are suitable for ex situ conservation in strain collections, which is the basis for future mass cultivation and commercial use.
