In the first stage of our research, C. officinalis and C. arvensis plants having medicinal and ec... more In the first stage of our research, C. officinalis and C. arvensis plants having medicinal and economical important were grown from embryo in the MS0 nutrient medium. In the second stage, callus culture was started with leaf explants of C. officinalis and C. arvensis plants and optimized with measuring of biomass in the MS0, MS1, MS3, MS4, MS6 nutrient mediums which were supplemented with combination of different concentrations of auxin and cytokinin. The statistical analysis relating to this measurements of callus biomass was conducted. The most favorable nutrient medium was established to be MS1 (1mg/l NAA+1mg/l BAP) for both C. officinalis and C. arvensis species during four different subculture from the 0. day. The most effective MS nutrient medium was determined to be MS1, MS4, MS3, MS6 and MS0, respectively.

The retention time of quercetin (6.445 min), kaempferol (11.246 min), caffeic acid (2.333 min), beta carotene (9.614 min) standards, which were planned to be analyzed by HPLC, was determined. The HPLC conditions that are most suitable for these metabolites have been determined. The amounts of quercetin, kaempferol, caffeic acid and beta carotene in the seedling leaves of these two Calendula species on the 0., 15., 30., 45., 60. days were compared. Distilled water (negative control), dimethyl sulfoxide (DMSO), ethyl methane sulfonate (EMS, positive control), 5% and 10% doses of V. opulus and E. arvense extracts were applied for 24 and 48 hours. Cytotoxicity and genotoxicity were evaluated by calculating %mitotic index and %chromosomal aberration index, respectively. When the 24h and 48h application of V. opulus and E. arvense extracts were compared with distilled water and DMSO applications; It was determined that these extracts increased the mitotic index and decreased the chromosomal aberration index. It was found that the mitotic index increased and the chromosomal aberration index decreased depending on the increase in the extract dose for 24h and 48h except for V. opulus. This research was to evaluate the effects of dodine on the physiology of Vicia faba L. through to... more This research was to evaluate the effects of dodine on the physiology of Vicia faba L. through total protein content and peroxidase (POX) activity . According to this, four weeks old seedlings were treated with 0.04 mL/L, 0.08 mL/L and 0.16 mL/L concentrations of dodine. Then, the leaves of plant samples were harvested with 24, 48 and 72 hours intervals. Our results have showed that while the total protein content were decreased significantly, POX activity was increased with the increased concentration of dodine and exposure time when compared with control seedlings. This research have shown that dodine can stimulate the plant defense system.
