INTRODUCTION
Immuno Assays
Immunoassays are the biological tests measure the concentration of substance in a biological liquid
typically serum or urine using immunological reaction of an antibody or anti antibodies to its antigen.
Radio immunoassay (RIA)
Radioimmunoassay (RIA) is a highly sensitive in vitro assay technique used to measure concentrations of
substances, usually measuring antigen concentrations (for example, hormone levels in blood) by use of
antibodies.

PRINCIPLE
The three key principles involved in RIA are:
1) Immune reaction
2) Competitive binding or competitive displacement reaction
3) Measurement of radio emission

Procedure
Take a micro tighter plate precoated with antibody
Add a radio labeled antigen (Hot antigen)
The hot antigen will bind with pre coated antibody
Remove the unbound hot antigen by washing by buffered solution
Add a patient sample which contain cold antigen (unlabeled antigen)
This cold antigen will compete with radio labeled antigen and replace
them
This replacement of cold antigen release them away from the antibody

CONCLUSION
SUMMARY
•RIA is an in-vitro assay technique used for the determination of Antigen concentration
present in a given sample.
•RIA have been successfully applied to a wide variety of pharmacological agents.
•RIA is an effective, precise and accurate method of quantifying concentrations of an antigen.
