EXPERIMENT NO. 1

Aim: To prepare a temporary mount of a leaf peel to show stomata.

Apparatus and Materials Required:

Calotropis leaf, forceps, watch glasses, glass slides, a dropper,


coverslips, blotting paper, safranin and a compound microscope.

Theory:

Stomata are pores found on the surface of leaves and young stems.
Stomata regulate the exchange of gases and water vapor
between the atmosphere and leaves.

Procedure -
1. Take a petunia leaf.
2. Remove a part of the peel from the lower surface of the
leaf. Keep the peels in a watch glass containing water.
3. Put a few drops of safranin stain in a watch glass.

4. After one minute take out the peel and place it on a clean glass
slide.
5. Put a drop of water over the peel and place a clean cover slip
gently over it.
6. Remove the excess stain with the help of blotting paper
7. Observe the slide under the compound microscope.

Observations:

1. Many small pores are seen scattered.


2. Each pore is guarded by two bean-shaped guard cells.
3. The stomata may be closed or opened .

Result:

Stomata are present on the surface of the leaf.

EXPERIMENT NO. 2

Aim: To find out the given substances as acids, bases and neutral substances and classify them.

Materials required:
Different samples, blue litmus and red litmus papers.

Procedure:

Take the samples and test them with blue and red LITMUS PAPER

1. Observe the changes and list out the changes in the table.

Observations:

The following observations are made:

Some of the solutions turn blue litmus to red litmus. Some of them turn red litmus to blue litmus. But
some solutions did not bring any change in litmus paper.

Inference:

1. The solutions which turn the blue litmus into red litmus are acidic substances
2. The solutions which turn the red litmus to blue litmus are basic substances .

3. But the solutions which do not change colour of any litmus paper are neutral substances.

4. From the above table acids, bases and neutral substances can be classified as follows.
