Packed Cell Volume (PCV) or
Haematocrit (HCT) Value
Introduction and principle:
Whole blood is centrifuged for maximum [Link] packing. The space occupied by
the [Link] is measured and expressed as a percentage of the whole blood volume.
Objectives:
To determine the volume or the amount of the [Link] in 100 ml (dl) of blood.

It requires less blood and also less time to determine a haematocrit.



— Materials And instruments
— 1-Microhaematocrit tube contain heparin and show a red ring at
the end of the tube
— 2-Microhaematocrit centrifuge
— 3. Plastic seal
— 4-Microhaematocrit reader

Procedure

1-Place the capillary tube in the blood sample allowing blood to enter the tube by

capillary action. Leave the last 15mm unfilled.

2-Seal the tube with modeling clay. Make sure there is no air trapped between the

clay and the column of blood.

3-Place the tube in the microhematocrit centrifuge with the sealed end to the outer

rim and centrifuge at 12000g for 5min.

4-Read the result using a microhematocrit reading device.

Results:

Normal ranges:

Adult males = 45 - 54%.


Adult females = 36 - 47%.
Children= 40-45%
Infants= 54-64%
