Information about predator–prey relationships in aquatic systems can be
helpful to improve the conservation management of endangered species.
Restoration efforts for the endangered axolotl (Ambystoma mexicanum)
involve the creation of refuges to increase the occurrence of suitable conditions
for native species. Little is known about the predator–prey interaction
between the axolotl and the native crayfish (Cambarellus montezumae) that
also inhabits the refuges. To understand this interaction, we designed a set of
experimental arenas and investigated both species at various life stages. Our
findings suggest a switch in the predator–prey relationship between crayfish
and axolotls at lengths between 2.6 and 3.5 cm. Individuals of both species
below this size-range serve as prey, whereas larger individuals become predators.
The 66.7% of crayfish that were preyed upon by axolotls were of intermediate
size, whereas adult crayfish primarily consumed newly hatched
axolotls (70%). We discuss the implications of this native species interaction
with respect to their habitat conservation.

Neural crest cells from both white mutant and dark (wildtype) axolotls (Ambystoma mexicanum) were cultured in increasing concentrations of fetal bovine serum (FBS; 2 to 20%). For each explant, the total number of cells that migrated and the percent of differentiated melanophores were recorded. At concentrations of FBS above 2% melanophore differentiation was essentially equivalent (32 to 59%) for both the white and dark neural crest cultures, but subtle differences in cell behavior and differentiation were found between the two phenotypes. By contrast there was a significant difference in the percent melanization of cells in serum-free control cultures, wherein melanophore differentiation in dark neural crest cultures was, on average, 18% compared to 5% in white cultures. Thus, contrary to all previously published work, white and dark neural crest cells are not intrinsically equivalent. Our culture results are discussed with regard to the probable in vivo conditions that cause the white phenotype.

Purpose Vision is encoded at photoreceptor synapses by the number of released vesicles and size of the post-synaptic response. We hypothesized that elevating cytosolic glutamate could enhance quantal size by increasing glutamate in vesicles. Methods We introduced glutamate (10–40 mM) into cone terminals through a patch pipette and recorded excitatory post-synaptic currents (EPSCs) from horizontal or OFF bipolar cells in the Ambystoma tigrinum retinal slice preparation. Results Elevating cytosolic glutamate in cone terminals enhanced EPSCs as well as quantal miniature EPSCs (mEPSCs). Enhancement was prevented by inhibiting vesicular glutamate transport with 1S,3R-1-aminocyclopentane-1,3-dicarboxylate in the patch pipette. How germ cells are specified in the embryos of animals has been a mystery for decades. Unlike most developmental processes, which are highly conserved, embryos specify germ cells in very different ways. Curiously, in mouse embryos germ cells are specified by extracellular signals; they are not autonomously specified by maternal germ cell determinants (germ plasm), as are the germ cells in most animal model systems. We have developed the axolotl (Ambystoma mexicanum), a salamander, as an experimental system, because classic experiments have shown that the germ cells in this species are induced by extracellular signals in the absence of germ plasm. Here, we provide evidence that the germ cells in axolotls arise from naive mesoderm in response to simple inducing agents. Solitary rod inner segments were isolated from salamander retinae. Their Ca current was studied with the 'whole-cell, gigaseal' technique (Hamill, Marty, Neher, Sakmann & Sigworth, 1981). The soluble constituents of the cytoplasm exchanged with the solution in the pipette. The external solution could be changed during continuous perfusion. Membrane voltage was controlled with a voltage clamp. After permeant ions other than Ca were replaced with impermeant ions (i.e. tetraethylammonium as a cation, and aspartate or methanesulphonate as an anion), an inward current remained. It activated at approximately -40 mV, reached a maximum at approximately 0 mV, and decreased as the membrane was further depolarized. The size of the current increased when Ba was substituted for external Ca. The current was blocked when Ca was replaced with Co. Purpose Efficient and precise release of glutamate from retinal bipolar cells is ensured by the positioning of L-type Ca2+ channels close to release sites at the base of the synaptic ribbon. We investigated whether Ca2+ channels at bipolar cell ribbon synapses are fixed in position or capable of moving in the membrane. Methods We tracked the movements of individual L-type Ca2+ channels in bipolar cell terminals after labeling channels with quantum dots (QDs) attached to α2δ4 accessory Ca2+ channel subunits via intermediary antibodies. Results We found that individual Ca2+ channels moved within a confined domain of 0.13–0.15 μm2 in bipolar cell terminals, similar to ultrastructural estimates of the surface area of the active zone beneath the ribbon. The hydroperiod of ephemeral wetlands is often the most important characteristic determining amphibian breeding success, especially for species with long development times. In mesic and wet pine flatwoods of the southeastern United States, ephemeral wetlands were a common landscape feature. Reticulated flatwoods salamanders (Ambystoma bishopi), a federally endangered species, depend exclusively on ephemeral wetlands and require at least 11 weeks to successfully metamorphose into terrestrial adults. We empirically modeled hydroperiod of 17 A. bishopi breeding wetlands by combining downscaled historical climate-model data with a recent 9-year record (2006-2014) of observed water levels. Empirical models were subsequently used to reconstruct wetland hydrologic conditions from 1896-2014 using the downscaled historical climate datasets. A reduction in extracellular chloride suppresses light-evoked currents of second-order retinal neurons (bipolar and horizontal cells) by reducing release of glutamate from photoreceptors. The underlying mechanisms responsible for this action of reduced extracellular Cl- were studied with a combination of electrophysiological recordings from single neurons in a retinal slice preparation and image analyses of intracellular Ca2+ (Fura-2) and pH [2',7'-bis-(2-carboxyethyl)-5-(and-6)-carboxyfluorescein, acetoxymethyl ester] in dissociated photoreceptors. The results show that reducing extracellular Cl- suppresses a dihydropyridine (DHP)-sensitive Ca2+ current (I(Ca)) in photoreceptors. It is proposed that suppression of I(Ca) results in suppression of photoreceptor neurotransmission. The suppressive effect of low Cl- on I(Ca) is not due to antagonism by the substituting anion nor is it mediated by changes in extracellular or intracellular pH.
