The potential coconut pulp protein can be processed in protein concentrate which has many benefits. This research was conducted to determine the effect of NaOH and (NH4)2SO4 concentrations in isolating protein and producing protein concentrates that have the highest yield and protein content. Isolation of coconut pulp protein was carried out using NaOH at several concentrations, namely 0, 0.2, 0.4, 0.6, 0.8, and 1 M, while the protein concentrate was prepared using the salting out method with a saturation level of the ammonium sulfate salt of 55%, 60%, and 65%. Analysis of protein content was carried out using the Kjeldahl method. The use of NaOH 0.2 M and (NH4)2SO4 with a saturation level of 60% was the chosen treatment with crude protein content and yield of 75.63% and 33.83%, respectively.

In this research, banana peel as adsorbent was carbonized at 350℃. Then chemically activated with ZnCl2 and KMnO4. Biopolymer pellets without banana peel activated carbon were in the form of white gel-like beads, activated carbon ZnCl2 and KMnO4 0.5 g were dark gray, and ZnCl2 and KMnO4 1 g were light gray. SEM results showed that the pores of ZnCl2 and KMnO4 1 g were larger than the pores without 0.5 g. The mechanical strength with the best percentage of activated ZnCl2 biopolymer pellets was 1 g at 6%, while activated KMnO4 was 0,5 g at 2%. Biopolymer pellets of banana peel activated ZnCl2 worked well in the removal of copper (II) at pH 6 the concentration absorbed 3.03 ppm. Meanwhile activated KMnO4 at pH 5 with the concentration absorbed 3.675 ppm.

Research on the antibacterial activity of ethanol extract from seedcoat of local Sorghum (Sorghum... more Research on the antibacterial activity of ethanol extract from seedcoat of local Sorghum (Sorghum bicolor [L.] Moench) against Staphylococcus aureus and Escherichia coli has been conducted. The purpose of this study was to determine the minimum inhibitory concentration of extract against both gram-positive and negative bacteria. The extraction method used in this study was a maceration method using ethanol as a solvent. Antibacterial activity test was carried out with several concentrations, namely 1 g/mL, 0.8 g/mL, 0.6 g/mL, 0.4 g/mL, 0.2 g/mL, 0.1 g/mL, 0.05 g/mL, 0.025 g/mL, and 0.01 g/mL. The results showed that the extract inhibited bacterial growth in a wide range of concentrations. The smallest inhibition was at a concentration of 0.05 g/mL with a clear zone diameter of 10.29 mm in Staphylococcus aureus and 11.70 mm in Escherichia coli. Ethanol extract of local Sorghum seedcoat exhibited a strong inhibition ability and probably a potential antibacterial agent source.

The study of the potential of Gempol wood stem extract (Nauclea orientalis L.) with three solvent... more The study of the potential of Gempol wood stem extract (Nauclea orientalis L.) with three solvent polarities has been exercised to determine the extract with a certain polarity and the smallest concentration that can inhibit the growth of bacteria Staphylococcus aureus and Escherichia coil. Extraction is performed using the maceration method with n-hexane, ethyl acetate, and ethanol as solvents. Furthermore, the potency in inhibiting the growth of bacteria was observed by the diffusion method with a range of extract concentrations from the largest to the smallest. The extract's ability to inhibit bacterial growth in the time range of up to 24 hours was measured using the turbidimetric method. The results showed that ethyl acetate and ethanol extracts were able to inhibit the growth of both bacteria in a wide concentration range (100 %– 5%), although n-hexane extracts showed the highest percentage in inhibiting both bacteria (15.27 mm and 16.89 mm, respectively). The purpose of this study was to determine the optimum condition in yielding crude amylase enzyme... more The purpose of this study was to determine the optimum condition in yielding crude amylase enzymes with the highest activity produced by PL-16, a thermophilic bacteria isolated from Pulu hot spring. The study was carried out using a completely randomized design with a varied incubation time of 0, 6, 12,18, 24, 30, 36, 42, 48, 54, 60, 66, and 72 hours, and agitation speed of 60, 90, 120, 150, 180, and 200 rpm. The activity of extracted crude amylase enzyme was determined using the DNS method and bacterial growth by measuring its Optical Density (OD600). The results showed that optimum amylase activity was at 48 hours incubation time with OD of 2.823 and activity of 0.437 U/mL. Furthermore, agitation speed of 180 rpm produced the highest amylase activity of 0.498 U/mL with an OD of 2.435. Tofu dregs are one of the waste products from making tofu that still has the potential to be proc... more Tofu dregs are one of the waste products from making tofu that still has the potential to be processed, because the protein content is relatively high, namely 23.62%. This waste has not been optimally utilized by the community unless it is used as animal feed. One alternative to utilizing tofu waste is processing it into specific products, namely producing protein hydrolysate using bromelain. The purpose of this study was to determine the best concentration of crude bromelain enzyme extract and hydrolysis time to obtain the degree of hydrolysis (DH) of protein hydrolysate in tofu dregs. DH of protein hydrolysate was determined with various concentrations of bromelain (8, 9, 10, 11, and 12%) (v/v) and variations in the hydrolysis time (120, 180, 240, and 300 minutes). The best degree of hydrolysis was 22.82% using 11% crude bromelain extract with a hydrolysis time of 120 minutes.

B1211 bacterial isolates with the highest Chitinolytic Index values of 1.71 were used in crude chitosanase production. The crude chitosanase production was performed using a Completely Randomized Design (RAL) consisting of four independent variables, in the form of chitosan substrate concentration (0.25%, 0.50%, 0.75%, 1.00%), agitation speed (60, 90, 120, and 150 rpm), fermentation time (1, 3, 5, 7, and 9 days), and fermentation temperature (30, 40, 50, and 60°C). The activity of chitosanase as dependent variable was determined by Schales method, which referred to standard curve of D-glucosamine. A neolignan, vladinol F, was successfully isolated from the dissolved fraction in acetone-ether of Dryobalanops oblongifolia stem bark. The chemical structure's determination was done based on an analysis of UV-Vis, NMR, and MS spectra and a comparison with the reference. In vitro test of this compound against Plasmodium falciparum 3D7 showed that IC50 value was 3.51 μg/mL.

They are classified based on their mechanism of actions in hydrolyzing starch. Amylases can be isolated from soils, plants, fungi, and bacteria, including thermophilic bacteria. The present study was focused on the determination of best temperature and pH of thermophilic amylase produced by thermophilic bacteria, isolated from Pulu Hotspring. As much as 30 isolates showed amylolytic activity in medium enriched with 1% starch. Of 3 isolates with the highest amylolytic index, PL 16 isolates with amylolytic index of 4.04 mm were studied further to find out its activity in amylose degradation. Enzyme activity was measured with the DNS method using maltose as standard. Temperature and pH measurement were varied in the range of 50, 60, 70, 80, 90, 100°C and 5, 6, 7, 8, 9, respectively. Research on the synthesis of gold nanoparticles using Muntingia calabura L. leaf extract as a bio... more Research on the synthesis of gold nanoparticles using Muntingia calabura L. leaf extract as a bioreductor and its potential analysis as a blood sugar nanosensor has been done. The synthesis Au nanoparticles are characterized and applied as sensors to detect glucose. The results show that gold nanoparticles have been successfully synthesized with a maximum wavelength of 544 nm. The measurements with PSA show that the average size of the gold nanoparticles is 78.2 nm. The gold nanoparticle-based glucose sensor design has a measurement range of 1-4 mM with a regression is 0.9248, minimum detection limit of the sensor at a concentration of 0.1500 mM and a maximum detection limit is 4.1744 mM with sensitivity is 0.6837 A. mM−1.mm−2.

RbAp46/48, histone chaperone, is a family of evolutionarily conserved WD40 repeat-containing prot... more RbAp46/48, histone chaperone, is a family of evolutionarily conserved WD40 repeat-containing proteins, which are involved in various chromatin-metabolizing processes, but their in vivo functional relevance is yet unclear. In order to examine the biological role of pRbAp48 in chicken DT40 cells, we generated a tetracycline-inducible system for conditional RbAp48-knockout cells. Depletion of RbAp48 led to delayed S phase progression associated with slow DNA synthesis and nascent nucleosome formation, followed by accumulation in G2/M phase, finally leading to cell death. Prior to cell death, these cells exhibited aberrant mitosis such as highly condensed and abnormal chromosome alignment on the metaphase plate, leading to chromosome missegregation. Depletion of RbAp48 also caused dissociation of heterochromatin protein 1 (HP1) from pericentromeric heterochromatin. Furthermore, depletion of RbAp48 from cells led to elevated levels of acetylation and slightly decreased levels of methylation, specifically at Lys-9 residue of histone H3. These results suggest that RbAp48 plays an important role in chromosome stability for proper organization of heterochromatin structure through the regulation of epigenetic mark.

Glucoamylase from the yeast Saccharomycopsis fibuligera R64 (GLL1) has successfully been purified... more Glucoamylase from the yeast Saccharomycopsis fibuligera R64 (GLL1) has successfully been purified and characterized. The molecular mass of the enzyme was 56,583 Da as determined by mass spectrometry. The purified enzyme demonstrated optimum activity in the pH range of 5.6–6.4 and at 50°C. The activity of the enzyme was inhibited by acarbose with the IC50 value of 5 μM. GLL1 shares high amino acid sequence identity with GLU1 and GLA1, which are Saccharomycopsis fibuligera glucoamylases from the strains HUT7212 and KZ, respectively. The properties of GLL1, however, resemble that of GLU1. The elucidation of the primary structure of GLL1 contributes to the explanation of this finding.

ER-stress-induced pre-emptive quality control (ER pQC) selectively degrades ER proteins via translocational attenuation during ER stress. However, the molecular mechanism underlying this process remains unclear. Here, we find that most newly synthesized endogenous transthyretin proteins are rerouted to the cytosol without cleavage of the signal peptide, resulting in proteasomal degradation in hepatocytes during ER stress. Derlin family proteins (Derlins), which are ER-associated degradation components, reroute specific ER proteins, but not ER chaperones, from the translocon to the proteasome through interactions with the signal recognition particle (SRP). Moreover, the cytosolic chaperone Bag6 and the AAA-ATPase p97 contribute to the degradation of ER pQC substrates.
