Pathological angiogenesis contributes directly to profound loss of II vision associated with many... more Pathological angiogenesis contributes directly to profound loss of II vision associated with many diseases of the eye. Recent work er suggests that human tyrosyland tryptophanyl-tRNA synthetases tin (TrpRS) link protein synthesis to signal transduction pathways in including angiogenesis. In this study, we show that a recombinant T: form of a COOH-terminal fragment of TrpRS is a potent antagonist la of vascular endothelial growth factor-induced angiogenesis in a la mouse model and of naturally occurring retinal angiogenesis in the et neonatal mouse. The angiostatic activity is dose-dependent in both ge systems. The recombinant fragment is similar in size to one genle erated naturally by alternative splicing and can be produced by e) proteolysis of the full-length protein. In contrast, the full-length N protein is inactive as an antagonist of angiogenesis. These results suggest that fragments of TrpRS, as naturally occurring and poT: tentially nonimmunogenic anti-angiogenics, can be used for the T: treatment of neovatcular eye diseases. te

The retina consists of organized layers of photoreceptors, interneurons, glia, epithelial cells, ... more The retina consists of organized layers of photoreceptors, interneurons, glia, epithelial cells, and endothelial cells. The economic model of supply and demand used to appropriately determine cost is highly applicable to the retina, in which the extreme metabolic demands of phototransduction are met by precisely localized and designed vascular networks. Proper development and maintenance of these networks is critical to normal visual function; dysregulation is characteristic of several devastating human diseases, including but not limited to age-related macular degeneration and diabetic retinopathy. In this article, we focus on the lessons learned from the study of retinal vascular development and how these lessons can be used to better maintain adult vascular networks and prevent retinal diseases. The present invention provides compositions and methods of treating neovascular diseases, such as... more The present invention provides compositions and methods of treating neovascular diseases, such as a retinal neovascular diseases and tumors, by administering to a patient suffering from a neovascular disease or tumor a vascular development inhibiting amount of a combination of the angiogenesis suppressing drugs comprising an angiostatic fragment of tryptophanyl-tRNA synthetase (TrpRS) and at least one compound selected from the group consisting of a vascular endothelial growth factor (VEGF) signaling inhibitor and an integrin signaling inhibitor. Compositions for use in the methods include an admixture of an angiostatic fragment of tryptophanyl-tRNA synthetase (TrpRS) and at least one of a vascular endothelial growth factor (VEGF) signaling inhibitor and an integrin signaling inhibitor, together with a pharmaceutically acceptable excipient.
