- Ethyl 3-amino-2-phenylpropanoate hydrochloride (4.38 g, 19.13 mmol) was suspended in methylene chloride (80 mL) and triethylamine (5.86 mL, 2.2 eq.). The mixture was cooled to 0°C and treated dropwise with ethyl 3-chloro-3-oxopropanoate (2.69 mL, 1.1 eq.). The reaction was kept at room temperature for one hour, diluted with methylene chloride, washed with 1 N HCI and then with 5% NaHC0 3 , dried over Na 2 S0 4 and evaporated to dryness.
- Example 14 2-(2-Amino-pyrimidin-4-yl)-3-iodo-1,5,6,7-tetrahydro-pyrrolo[3,2-c]pyridin-4-one KOH (61 mg, 1.09 mmol) was added to a solution of 2-(2-amino-pyrlmidin-4-yl)-1 , 5,6,7- tertrahydro-pyrrolo[3,2-c]pyridin-4-one (100 mg, 0.44 mmol) in DMF (5 ml). A solution of iodine (115 mg, 0.45 mmol) in DMF (2 ml) was added . After 30 min the reaction mixture was poured onto ice water (containing 0.5 ml NH 3 and 25 mg K 2 S 2 0 5 ). The yellow precipitate was filtered, washed with cold water and dried. The compound was purified by flash chromatography
