Abstract
Description
도 2는 상기 벡터로 형질전환된 균주로부터 효소 단백질을 정제한 후SDS-PAGE를 통하여 단량체의 크기를 확인한 결과를 나타낸 것이다.
도 3은 본 발명의 D-사이코스 3-에피머화 효소 단백질의 온도에 따른 활성(과당 기질로부터 사이코스의 전환율)을 나타낸 것이다.
도 4는 본 발명의 D-사이코스 3-에피머화 효소 단백질의 pH에 따른 활성(과당 기질로부터 사이코스의 전환율)을 나타낸 것이다.
Claims (12)
- 서열번호 1의 아미노산 서열로 이루어진 단백질 또는 상기 단백질을 발현하는 균주를 포함하는, 과당으로부터 사이코스의 제조용 조성물.
- 제1항에 있어서, 상기 단백질은 과당의 3번 탄소 위치를 에피머화하여 사이코스로 전환하는 것인 조성물.
- 제1항에 있어서, 상기 서열번호 1의 아미노산 서열로 이루어진 단백질은 루미노코코스 토르크 (Ruminococcus torques) 로부터 유래된 효소 단백질인 조성물.
- 제1항에 있어서, 상기 서열번호 1의 아미노산 서열로 이루어진 단백질은 서열번호 2 또는 서열번호 3의 염기 서열로 이루어진 유전자에 의해 코딩되는 것인 조성물.
- 제1항에 있어서, 상기 균주는 서열번호 1의 아미노산 서열로 이루어진 단백질을 코딩하는 유전자를 포함하는 재조합 벡터로 형질전환된 것인 조성물.
- 제5항에 있어서, 상기 유전자는 서열번호 2 또는 서열번호 3의 염기 서열로 이루어진 것인 조성물.
- 제5항에 있어서, 상기 재조합 벡터는 도 1의 개열 지도를 가지는 재조합 벡터인 조성물.
- 제1항에 있어서, 상기 단백질은 다음의 특징을 가지는 것인 조성물 :
(a) 분자량이 32 내지 34 kDa;
(b) 최적 활성 온도가 50 내지 65℃; 및
(c) 최적 활성 pH가 pH7.0 내지 9.0. - 제1항 내지 제8항 중 어느 한 항의 조성물을 과당과 반응시키는 단계를 포함하는, 과당으로부터 사이코스를 제조하는 방법.
- 제9항에 있어서, 상기 반응은 50 내지 65℃ 에서 이루어지는 것인 방법.
- 제9항에 있어서, 상기 반응은 pH7.0 내지 9.0 에서 이루어지는 것인 방법.
- 제9항에 있어서, 상기 과당은 5 내지 60%(w/v)의 농도인 방법.
