Claims (6)

1. Förfarande för framställning av väsentligen rent oglykosy-lerat humant interleukin-2-protein med en specifik aktivitet om minst 10“ U/mg och en renhet om minst 99 %, soin uppvisar ett enda band vid analys med SDS-polyakrylamidelektrofores under saväl reducerande som icke-reducerande betingelser och som är väsentligen fritt frän endotoxin och pyrogen, k ä n -netecknat av, att man odlar en transformant av Escherichia coli, som innehäller en DNA med en bassekvens som kodar för humant interleukin-2 sä, att man ästadkommer produktion och ackumulation av humant interleukin-2 i odlingsmediet, och underkastar den sä erhällna humant interleukin-2-inne-hällande lösningen ett reningsförfarande som utnyttjar HPL-kromatografi med en revers-fas kolonn.
2. Förfarande enligt patentkravet 1, kännetecknat av, att kromatografin utförs under användande av en eluent med pH 1, 5-8.
3. Förfarande enligt patentkravet 1, kännetecknat av, att kromatografin utförs med strömningshastigheten 0, 1-100 ml / min.
4. Förfarande enligt patentkravet 1, kännetecknat av, att humant interleukin-2 produceras och ackumuleras i växande trans formantceller och att det humana interleukinet-2 extraheras ur cellerna, varvid man erhäller en vätska som innehäller humant interleukin-2.
5. Förfarande enligt patentkravet 4, kännetecknat av, att det humana interleukinet-2 extraheras med en lösning som innehäller proteindenaturerande ämne.
6. Förfarande enligt patentkravet 5, kännetecknat av, att det humana interleukinet-2 extraheras med en lösning av ett guanidinsalt med en koncentration om 2-8 M.
