The two medicines have opposite effects, and are generally applied less simultaneously in the traditional anti-alcoholism composition, but the applicant has found in clinical practice that the 2 kinds of medicines are used together, and are matched with the leaves of the Ampelopsis (Ampelopsis Michx.) plant of the family Vitaceae, and the digestant has unexpected curative effect.
One aspect of the present invention relates to a composition comprising: the leaf, digestant, interior-warming and heat-clearing herbs of Ampelopsis genus (Ampelopsis Michx) of Vitaceae family, or their extracts or effective components.
The composition is applied to the preparation of medicines, health-care foods and foods for preventing and relieving acute alcoholism or drunkenness, wherein the symptoms of acute alcoholism or drunkenness comprise: headache, dizziness, hypodynamia, vomiting, accelerated heartbeat, stomachache, inattention and other symptoms; the symptoms begin to improve within 10-15 minutes after the composition is administered.
The Ampelopsis plant of Vitaceae is Ampelopsis grossedentata (A. megalophylla Diels et. Gilg), Ampelopsis grossedentata (hand. Mazz.) W.T.Wang, Ampelopsis grossedentata [ A.sincia (Miq.) W.T.Wang ], Ampelopsis grossedentata [ A.sincisvar.hancei (pl.) W.T.Wan, Ampelopsis heterophylla [ A.humulifolia.var.Heterophylla (Thunb.) K.Koch ], Ampelopsis trisplifaensis [ A.delavayana pl. ] or Myrgle ampelopsis grossedentata [ A.breviedaphala-var.kularensis Red. ]; the digestant is hawthorn or dark plum; the interior warming medicine is cinnamon, cassia twig, dried ginger, common fennel, clove, pepper or piper longi; the heat-clearing medicine is fructus Gardeniae, Gypsum Fibrosum, herba Taraxaci, herba Lobeliae chinensis, herba Houttuyniae, rhizoma Smilacis Glabrae, herba Hedyotidis Diffusae, Trichosanthis radix or semen Cassiae.
Preferably, the raw materials of the composition comprise: ampelopsis grossedentata leaf or Ampelopsis grossedentata leaf, fructus crataegi or mume fructus, cortex Cinnamomi or fructus Piperis, and fructus Gardeniae or herba Taraxaci; more preferably, the raw materials of the composition comprise: ampelopsis grossedentata leaf, hawthorn, cinnamon and gardenia.
The pharmaceutical composition of any one of the present invention comprises the following raw material medicines by weight: 50-150 parts of Ampelopsis grossedentata leaf or Ampelopsis grossedentata leaf, 50-150 parts of hawthorn or dark plum fruit, 5-30 parts of cinnamon or pepper, and 10-70 parts of gardenia or dandelion; preferably, 70-130 parts of Ampelopsis grossedentata leaf or Ampelopsis grossedentata leaf, 70-130 parts of hawthorn or dark plum, 10-25 parts of cinnamon or pepper, and 20-60 parts of cape jasmine or dandelion; more preferably, 90-110 parts of Ampelopsis grossedentata leaf or Ampelopsis grossedentata leaf, 90-110 parts of hawthorn or dark plum, 15-20 parts of cinnamon or pepper, and 30-50 parts of cape jasmine or dandelion.
The pharmaceutical composition of the invention can also be prepared by directly crushing the raw materials.
The raw material medicaments in the pharmaceutical composition can be replaced by corresponding extracts or effective components. For example, the composition adopts the following extracts in weight ratio as raw materials: 50-150 parts of ampelopsis grossedentata leaf ethanol extract, 50-150 parts of hawthorn ethanol extract, 5-30 parts of cinnamon ethanol extract and 10-70 parts of gardenia ethanol extract; preferably, the content of the ampelopsis grossedentata leaf ethanol extract is 70-130 parts, the content of the hawthorn ethanol extract is 70-130 parts, the content of the cinnamon ethanol extract is 10-25 parts, and the content of the gardenia ethanol extract is 20-60 parts; more preferably, 90-110 parts of ampelopsis grossedentata leaf ethanol extract, 90-110 parts of hawthorn ethanol extract, 15-20 parts of cinnamon ethanol extract and 30-50 parts of gardenia ethanol extract; most preferably, 100 parts of ampelopsis grossedentata leaf ethanol extract, 100 parts of hawthorn ethanol extract, 18 parts of cinnamon ethanol extract and 40 parts of gardenia ethanol extract.
Preferably, the total flavone content in the ampelopsis grossedentata leaf ethanol extract is not less than 30%, the total flavone content in the hawthorn ethanol extract is not less than 30%, the cinnamaldehyde content in the cinnamon ethanol extract is not less than 10%, and the geniposide content in the gardenia ethanol extract is not less than 30%; preferably, the total flavone content in the ampelopsis grossedentata leaf ethanol extract is not less than 50%, the total flavone content in the hawthorn ethanol extract is not less than 50%, the cinnamaldehyde content in the cinnamon ethanol extract is not less than 20%, and the geniposide content in the gardenia ethanol extract is not less than 50%. The above extracts or effective components can be prepared by the existing preparation process in the prior art, and also can be incorporated into the technical scheme of the invention, and the above extracts and monomers can also be purchased from commercial products.
For example, the Ampelopsis grossedentata ethanol extract, the Crataegus pinnatifida ethanol extract, the Cinnamomum cassia ethanol extract, the Gardenia jasminoides Ellis extract, the Scutellaria baicalensis extract, and the Taraxacum officinale extract can be prepared by respectively extracting with 30-90% ethanol, vacuum concentrating at 40-80 deg.C, drying at low temperature, and mixing at a certain proportion; or extracting with 30-90% alcohol respectively, separating with macroporous resin, vacuum concentrating at 40-80 deg.C, drying at low temperature, and mixing at a certain proportion.
The invention also provides a pharmaceutical preparation, which is any pharmaceutically acceptable dosage form prepared from the pharmaceutical composition and pharmaceutically acceptable auxiliary materials. The preparation can be conventional oral preparations such as common tablets, sustained-release tablets, orally disintegrating tablets, buccal tablets, granules, hard or soft capsules, solutions and the like.
The composition can also be widely used in medicines, health products and health foods.
The composition of the present invention contains an effective amount of the composition as an active ingredient and is formulated into various oral preparations. When formulated, the formulation may be carried out using conventional fillers, extenders, binders, wetting agents, disintegrants, diluents such as surfactants, or excipients. More preferably, the ethanol extract is prepared by respectively extracting with 60% ethanol, vacuum concentrating at 60 deg.C, drying at low temperature, and mixing at a certain proportion; or extracting with 60% ethanol respectively, separating with macroporous resin, vacuum concentrating at 60 deg.C, drying at low temperature, mixing at a certain proportion,
the Ampelopsis grossedentata leaves are new resource food (bulletin number: No. 16 in 2013) approved by the Ministry of health of China, contain a large amount of flavonoids, and have special effects on relieving alcoholism, preventing alcoholic liver and fatty liver and protecting liver. Cinnamon integrates edible and medicinal functions, has the effects of warming stomach, relieving diarrhea, stimulating the menstrual flow, expelling cold and dampness, promoting urination, reducing blood pressure, controlling blood sugar balance and the like, and also has certain functions of refreshing and relieving alcoholism. "Haw, a mountain fruit" is recorded in "follow-up family diet notation" of Qing-Wang Meng Ying. Bei Dai, also known as Biao Tuo, commonly known as hong Guo. Sour, sweet and warm. Detailed Description
Ampelopsis grossedentata leaf, Hawthorn, cinnamon, and cape jasmine were purchased from Nanjing Miichong drugstore.
Pulverizing the above materials respectively, and sieving with 80 mesh sieve.
Example 1
Respectively soaking the powder of Ampelopsis grossedentata leaf, hawthorn, cinnamon and gardenia in 60% ethanol at room temperature for 2 hours; extracting with reflux method (at 65 deg.C for 2 times, 4 hr each time, the first material-liquid ratio is 1: 10, and the second material-liquid ratio is 1: 6); carrying out vacuum filtration; mixing filtrates, vacuum concentrating, drying at low temperature, and making into extract with relative density of 1.08-1.10(60 deg.C).
Example 2
Taking the extract of example 1, and extracting according to the weight ratio of the ampelopsis grossedentata leaf: and (3) hawthorn extract: cinnamon extract: gardenia extract 100: 100: 18: 40, taking 100g of the mixture, 150g of starch, 12g of 10% starch slurry, 12g of dry starch and 1.5g of magnesium stearate, and preparing 300 tablets by a conventional tablet preparation method.
Example 3
Taking the extract of example 1, and extracting according to the weight ratio of the ampelopsis grossedentata leaf: cinnamon extract: gardenia extract 100: 18: 40, taking 100g of the mixture, 150g of starch, 12g of 10% starch slurry, 12g of dry starch and 1.5g of magnesium stearate, and preparing 300 tablets by a conventional tablet preparation method.
Example 4
Taking the extract of example 1, and extracting according to the weight ratio of the ampelopsis grossedentata leaf: and (3) hawthorn extract: cinnamon extract-100: 100: 18, taking 100g of the mixture, 150g of starch, 12g of 10% starch slurry, 12g of dry starch and 1.5g of magnesium stearate, and preparing 300 tablets by a conventional tablet preparation method.
Experimental example 1 toxicological test
The mixed extract of example 2 is administered to mice at 2500mg/kg daily by intragastric administration, and no obvious toxic symptoms or death is observed after one week of observation, which indicates that the pharmaceutical composition of the present invention has little acute toxicity when administered orally.
The mixed extract in the example 2 is administrated to rats by intragastric administration for 30 days and 90 days at 500mg/kg, 1000 mg/kg and 2000mg/kg daily, the diet, the body weight, the hematuria and the liver and kidney functions of the rats after long-term administration are not obviously changed, and the mixed extract has no obvious toxicity on pathological histological observation of 18 important organs and tissues of experimental animals, such as heart, brain, liver, kidney and the like.
4 normal people voluntarily take 1.5g or 3g of the mixed extract of the example 2 every day, and after one week, compared with the prior and the post-taking, the blood sugar, the blood fat and the blood pressure have no obvious change, and the blood routine and the urine routine have no obvious change.
Experimental example 2 animal experiments
Acute poisoning test of mice caused by ethanol
(1) The method comprises the following steps: 50 Kunming mice are taken, male and female halves are divided into 5 groups randomly, namely a blank control group and an ethanol model group, the composition of the invention is low (0.5g/kg), medium (2.5g/kg) and high dose group (5.0g/kg), and each group comprises 10 animals. The administration groups respectively irrigate different doses of the mixed extract liquid medicine of the invention 2, and irrigate equal-volume physiological saline of 20mL/kg for the blank control group and the model group. After 30min, the rest components are respectively gazed with 65 percent (13g/kg) of ethanol 20mL/kg except that the blank control group is given equal volume of distilled water. The number of deaths and survivals for each group of animals over 24h were then recorded.
(2) As a result: the blank control group of 10 mice was not infused with ethanol and none of the animals died. Most of 10 animals in the model group died within 18 hours after intragastric administration of 65% (13g/kg) ethanol, and the death rate reaches 80%, which is acute toxic reaction and death reaction of mice caused by ethanol. The mortality rates of 10 animals in the low-dosage group and the medium-dosage group are respectively 50% and 30%; in the high dose group, 1 animal died, with a mortality rate of 10%. The composition can reduce the death rate of mice caused by alcohol poisoning and has a protective effect on acute alcoholism.
Experimental example 3 summary of clinical trial data
60 healthy volunteers were selected, age 28-58. 10 volunteers were randomly selected as 1 group and divided into 6 groups.
The drunkenness prevention experiment:
the first group of volunteers took 3 tablets of the tablet of example 2 before drinking wine, and then took 100ml of red star 43 degree aged Erguotou per person, 10 persons did not have symptoms such as dizziness, vomiting, stomachache and the like.
The second group of volunteers took 3 tablets of example 3 before drinking wine, and then took 100ml of red star 43 degree aged Erguotou per person, 6 of 10 persons showed symptoms of dizziness.
The third group of volunteers took 3 tablets of example 3 before drinking wine, and then took 100ml of red star 43 degree aged Erguotou per person, and 5 out of 10 persons showed symptoms of dizziness.
An anti-alcohol experiment:
each of the four groups of volunteers took 100ml of red star 43 degree aged Erguotou, 8 of 10 people showed dizziness, of which 3 felt burning pain in the stomach, and after taking 3 tablets of the tablet of example 2, the latter was asked again after 15 minutes, 7 people all had no discomfort, 1 person had dizziness and was good, and the stomach was still slightly uncomfortable.
Each of the fifth group of volunteers took 100ml of red star 43 degree aged Erguotou, 9 of 10 people showed symptoms of dizziness, 2 of which felt burning pain in the stomach, and after taking 3 tablets of the tablet of example 2, the latter was asked again after 15 minutes, 3 people did not feel dizzy, 1 person did not feel uncomfortable in the stomach, and 5 other people showed no relief in symptoms.
Each of the sixth group of volunteers took 100ml of red star 43 degree aged Erguotou, 8 of 10 people showed symptoms of dizziness, 4 of them felt burning pain in the stomach, after taking 3 tablets of the tablet of example 2, the latter was asked again after 15 minutes, 2 people did not feel dizzy in the head, 2 people did not feel uncomfortable in the stomach, and 4 other people showed no relief in symptoms.
Typical cases are as follows:
1. zhangzhi, male, 54 years old, dizziness and stomachache after drinking 2 two white spirits at one time at ordinary times. After the composition is taken, 5 two white spirits are drunk at one time, the head is still clear, and discomfort of the stomach is avoided.
2. Some Guo, male, 36 years old, sells and mostly drinks at ordinary times, and after drunk, the drunkenness-relieving products such as Haiwang Jinzun and Jiuqian cup are taken, but the drunkenness symptom cannot be relieved. After one-time drunk, after the composition is taken, the feelings of dizziness, vomiting and liver area swelling are relieved within about 10 minutes, the head is recovered to be clear, the vomiting is disappeared, and the liver area is comfortable.

The pharmaceutical composition of claim 3, wherein: the composition comprises the following raw material medicines in parts by weight: 50-150 parts of Ampelopsis grossedentata leaf or Ampelopsis grossedentata leaf, 50-150 parts of hawthorn or dark plum fruit, 5-30 parts of cinnamon or pepper, and 10-70 parts of gardenia or dandelion; preferably, 70-130 parts of Ampelopsis grossedentata leaf or Ampelopsis grossedentata leaf, 70-130 parts of hawthorn or dark plum, 10-25 parts of cinnamon or pepper, and 20-60 parts of cape jasmine or dandelion; more preferably, 90-110 parts of Ampelopsis grossedentata leaf or Ampelopsis grossedentata leaf, 90-110 parts of hawthorn or dark plum, 15-20 parts of cinnamon or pepper, and 30-50 parts of gardenia or dandelion; most preferably, 100 parts of ampelopsis grossedentata leaves, 100 parts of hawthorn, 18 parts of cinnamon and 40 parts of gardenia.

8. Use of the composition of any one of claims 1 to 7 for the preparation of a medicament, health food, food for preventing and alleviating acute alcoholism or intoxication.
9. The use of claim 8, wherein the symptoms of acute alcoholism or intoxication comprise: headache, dizziness, hypodynamia, vomiting, accelerated heartbeat, stomachache, inattention and the like.
10. The use according to claim 9, wherein the symptoms begin to improve within 10 to 15 minutes after the composition is administered.
