Abstract
本发明涉及一种植物抗氧化组合物及其日化品的制备与应用。所述的植物抗氧化组合物包括如下重量份的组分:黄芩饮片1‑40份、白梅花饮片1‑40份、丹参饮片0.1‑20份、白薇饮片0.1‑20。本发明所述的组合物,均来源于天然抗氧化成分。经研究证明,该植物组合物对常见的DPPH自由基和ABTS自由基均有不同程度的清除效果。可制备具有对抗氧化损伤、延缓衰老、改善皮肤暗黄、减轻细纹、增加皮肤光泽度等功能的日化产品,包括面膜、精华水、护肤乳膏、护肤凝胶、洁面乳、沐浴乳等剂型,用于全身各部位的清洁和维护。

背景技术:
人类的生活节奏随着时间推移在飞速加快,同时,现代城市中空气污染也变得越来越严重。许多人在繁忙、高压的生活下养成了很多不良的生活习惯,导致越来越多的人的皮肤受到自由基的过度侵害,出现暗沉、敏感、过早衰老等一系列皮肤问题。目前,自由基损伤被认为是细胞老化、病变的主要因素,而皮肤,由于经常暴露于氧和紫外线中,是自由基造成的氧化应激损伤最大的人体组织。在已知的多种自由基中,最常见的是活性氧分子,它包括羟基自由基、过氧化氢、超氧化物和单氧。研究显示,氧自由基引起脂质氧化,产生对细胞膜的损伤,从而导致未老先衰和最终的细胞死亡。同时,多数研究者认为,活性氧对皮肤能产生紫外线诱导的损伤,也能导致粉刺、皱纹、老年斑、下垂、干燥甚至癌症。
自由基的清除剂被称为抗氧化剂,通过抑制或终止氧自由基链式反应而达到抗氧化作用。抗氧化剂对皮肤氧化应激损伤造成的皮肤老化、病变等,具有显著的预防和改善作用。如目前应用较多的超氧化物歧化酶(SOD)、维生素C、维生素E等。但此类物质多有不稳定现象,导致其抗氧化作用不能很好的发挥。近年来,随着人们对化妆品安全高效、无刺激等需求的增长,植物来源的抗氧化剂因其来源广泛、安全有效、天然无毒等特性被日益重视。
根据实验研究证明,黄芩能清热燥湿、泻火解毒,具有消除自由基、抗氧化、抗癌、抗病毒、抗菌、抗过敏、保护神经元等多方面的药理作用;白梅花具有清除氧自由基、防止黑色素沉积、抗血小板凝聚、抗抑郁等功效,能提亮肤色,令肌肤焕发青春活力;丹参能够有效抑制皮肤脂质氧化,改善局部微循环,消除由于血液循环不畅而导致的暗疮、色斑,调节水油平衡;白薇能清热凉血、利尿通淋、解毒疗疮的功效,具有退热、抗炎、抗肿瘤、抗氧化、改善妇女更年期症状等功效。
综上所述,随着人们对肌肤护理问题的重视程度增加,采用上述成分制备的植物抗氧化组合物及其日化品具有健康高效、绿色环保、实用价值高和市场前景广阔等优势。

分别取1-30份的肉豆蔻酸、月桂酸、硬脂酸和0.5-20份的甘油,加热融化,加入1-30份的氢氧化钾30-90℃搅拌5-180min。加入0.001-0.1份的植物抗氧化组合物、1-10份的丙二醇、0.1-10份的聚山梨醇酯-80加热混合,搅拌均匀后加入0.5-10份的椰油酰胺甜菜碱,0.5-10份的椰油酰两性基二乙酸二钠,0.5-10份的棕榈酸异丙酯,0.1-20份的二异硬脂醇苹果酸酯,0.5-10份的单硬脂酸甘油酯,0.5-10份的鲸蜡硬脂基葡糖苷,0.01-3份的聚季铵盐-39,0.01-2份的防腐剂,0.01-3份的香精和适量水,保温混合,冷却,灌装,即得。

1.提取:
分别取重量份为20份的黄芩、40份的白梅花饮片、20份的丹参饮片和0.1份的白薇饮片,混合粉碎,加50%乙醇回流提2次,每次加8倍量溶剂,回流120分钟,提取液合并,过滤,滤液浓缩,备用。
将上述浓缩液过大孔树脂柱,50%乙醇洗脱,洗脱液合并,干燥即得。
2.制备:
分别取重量份0.1份的植物抗氧化组合物、0.001份的虾青素,加入0.5份的二乙二醇单乙基醚、1份的聚山梨醇酯-80和5份的氢化卵磷脂混合均匀后,加入0.01份的水溶性神经酰胺、0.3份的卡波姆、0.05份的防腐剂、0.01份的香精和适量水,充分搅拌混合均匀,分装,即得。
实施例2:
一种植物抗氧化组合物的面膜液,该日化品的制备方法如下:
1.提取:
分别取重量份为40份的黄芩、20份的白梅花饮片、0.1份的丹参饮片和20份的白薇饮片,混合粉碎,加20%乙醇回流提3次,每次加3倍量溶剂,回流180分钟,提取液合并,过滤,滤液浓缩,备用。
将上述浓缩液过大孔树脂柱,20%乙醇洗脱,洗脱液合并,干燥即得。
2.制备:
分别取重量份0.001份的植物抗氧化组合物、0.1份的虾青素,0.1份的辅酶Q10,加入25份的二乙二醇单乙基醚、8份的聚山梨醇酯-80和0.05份的氢化卵磷脂混合均匀后,加入10份的紫皮石斛提取物、10份的灵芝提取物、0.3份的黄原胶、1份的防腐剂、0.06份的香精和适量水,充分搅拌混合均匀,分装,即得。

1.提取:
分别取重量份为1份的黄芩、40份的白梅花饮片、20份的丹参饮片和20份的白薇饮片,混合粉碎,加70%乙醇回流提2次,每次加15倍量溶剂,回流60分钟,提取液合并,过滤,滤液浓缩,备用。
将上述浓缩液过大孔树脂柱,70%乙醇洗脱,洗脱液合并,干燥即得。
2.制备:
分别取重量份2份的异构十二烷、2份的二异硬脂醇苹果酸酯、0.5份的聚二甲基硅氧烷、0.5份的聚甲基倍半硅氧烷、0.01份的虾青素、1份的乙二醇双硬脂酸酯、10份的乳木果油,加热混合均匀,加入2份的植物抗氧化组合物与5份的二乙二醇单乙基醚、2份的聚山梨醇酯-80的混合液,加热混合均匀,再加入0.1份的脯氨酸、0.1份的精氨酸、0.1份的缬氨酸、0.1份的苏氨酸、0.001份的透明质酸钠、5份的丙二醇、10份的单双硬脂酸甘油酯、10份的聚甘油-3双硬脂酸酯、5份的甘油聚乙二醇-75-硬脂酸酯、0.5份的防腐剂、1份的香精和适量水,保持85℃均质乳化45分钟,冷却,灌装,即得。
实施例4:
一种植物抗氧化组合物的护肤水,该日化品的制备方法如下:
1.提取:
分别取重量份为40份的黄芩、1份的白梅花饮片、20份的丹参饮片和20份的白薇饮片,混合粉碎,加90%乙醇回流提1次,每次加20倍量溶剂,回流30分钟,提取液合并,过滤,滤液浓缩,备用。
将上述浓缩液过大孔树脂柱,90%乙醇洗脱,洗脱液合并,干燥即得。
2.制备:
分别取重量份0.01份的植物抗氧化组合物、0.001份的虾青素、0.001份的辅酶Q10,加入10份的二乙二醇单乙基醚、1份的聚山梨醇酯-80和0.5份的氢化卵磷脂混合均匀后,加入0.001份的脯氨酸、0.001份的精氨酸、0.001份的缬氨酸、0.001份的苏氨酸、0.2份的聚丙烯酰胺、0.2份的防腐剂、0.3份的香精和适量水,充分搅拌混合均匀,灌装,即得。
实施例5:
一种植物抗氧化组合物的精华喷雾,该日化品的制备方法如下:
1.提取:
分别取重量份为20份的黄芩、20份的白梅花饮片、10份的丹参饮片和10份的白薇饮片,混合粉碎,加60%乙醇回流提2次,每次加10倍量溶剂,回流90分钟,提取液合并,过滤,滤液浓缩,备用。
将上述浓缩液过大孔树脂柱,60%乙醇洗脱,洗脱液合并,干燥即得。
2.制备:
分别取重量份10份的植物抗氧化组合物、0.01份的虾青素、0.01份的辅酶Q10,加入30份的二乙二醇单乙基醚、5份的聚山梨醇酯-80和2份的氢化卵磷脂、0.05份的乳木果油、2份的油酸聚乙二醇甘油酯混合均匀后,加入0.01份的脯氨酸、0.01份的精氨酸、0.01份的缬氨酸、0.01份的苏氨酸、1份的PEG-40氢化蓖麻油、0.05份的防腐剂、0.1份的香精和适量水,充分搅拌混合均匀,低温混合氮气,灌装,即得。
实施例6:
一种植物抗氧化组合物的洁面乳,该日化品的制备方法如下:
1.提取:
分别取重量份为30份的黄芩、30份的白梅花饮片、5份的丹参饮片和5份的白薇饮片,混合粉碎,加40%乙醇回流提3次,每次加6倍量溶剂,回流150分钟,提取液合并,过滤,滤液浓缩,备用。
将上述浓缩液过大孔树脂柱,40%乙醇洗脱,洗脱液合并,干燥即得。
2.制备:
分别取10份的肉豆蔻酸、月桂酸、硬脂酸和15份的甘油,加热融化,加入8份的氢氧化钾60℃搅拌90min。加入0.05份的植物抗氧化组合物、5份的丙二醇、0.5份的聚山梨醇酯-80加热混合,搅拌均匀后加入5份的椰油酰胺甜菜碱,5份的椰油酰两性基二乙酸二钠,5份的棕榈酸异丙酯,5份的二异硬脂醇苹果酸酯,5份的单硬脂酸甘油酯,5份的鲸蜡硬脂基葡糖苷,0.5份的聚季铵盐-39,0.02份的防腐剂,1份的香精和适量水,保温混合,冷却,灌装,即得。
实施例7:
一种植物抗氧化组合物的面膜液,该日化品的制备方法如下:
1.提取:
分别取重量份为1份的黄芩、40份的白梅花饮片、20份的丹参饮片和20份的白薇饮片,混合粉碎,加70%乙醇回流提2次,每次加15倍量溶剂,回流60分钟,提取液合并,过滤,滤液浓缩,备用。

一种植物抗氧化组合物的免洗面膜,该日化品的制备方法如下:
1.提取:
分别取重量份为30份的黄芩、30份的白梅花饮片、5份的丹参饮片和5份的白薇饮片,混合粉碎,加40%乙醇回流提3次,每次加6倍量溶剂,回流150分钟,提取液合并,过滤,滤液浓缩,备用。
将上述浓缩液过大孔树脂柱,40%乙醇洗脱,洗脱液合并,干燥即得。
2.制备:
分别取重量份0.5份的异构十二烷、0.5份的二异硬脂醇苹果酸酯、1份的聚二甲基硅氧烷、0.1份的聚甲基倍半硅氧烷、0.1份的虾青素、0.9份的乙二醇双硬脂酸酯、0.1份的乳木果油,加热混合均匀,加入1份的植物抗氧化组合物与1份的二乙二醇单乙基醚、0.5份的聚山梨醇酯-80的混合液,加热混合均匀,再加入0.01份的脯氨酸、0.01份的精氨酸、0.01份的缬氨酸、0.01份的苏氨酸、1份的透明质酸钠、0.5份的丙二醇、1份的单双硬脂酸甘油酯、5份的聚甘油-3双硬脂酸酯、10份的甘油聚乙二醇-75-硬脂酸酯、0.1份的防腐剂、0.1份的香精和适量水,保持75℃均质乳化60分钟,冷却,灌装,即得。
实施例9:
一种植物抗氧化组合物的面膜液,该日化品的制备方法如下:
1.提取:
分别取重量份为20份的黄芩、40份的白梅花饮片、20份的丹参饮片和0.1份的白薇饮片,混合粉碎,加50%乙醇回流提2次,每次加8倍量溶剂,回流120分钟,提取液合并,过滤,滤液浓缩,备用。
将上述浓缩液过大孔树脂柱,50%乙醇洗脱,洗脱液合并,干燥即得。
2.制备:
分别取重量份0.1份的植物抗氧化组合物、0.1份的虾青素,加入10份的二乙二醇单乙基醚、0.5份的聚山梨醇酯-80和1份的氢化卵磷脂混合均匀后,加入1份的水溶性神经酰胺、0.6份的卡波姆、0.1份的防腐剂、0.1份的香精和适量水,充分搅拌混合均匀,分装,即得。
实施例10:
一种植物抗氧化组合物的DPPH自由基清除和ABTS自由基清除实验:
1.实验方法:
1.1DPPH自由基清除实验
本实验采用吸光光度法进行。在96孔板内加入样品,设3个复孔,每个样品2个稀释梯度,进行10倍倍比稀释(初筛后若样品具有明显的DPPH自由基清作用,则每个样品4个稀释梯度,进行2倍倍比稀释进行复筛),每孔100μL,每孔加入0.1M DPPH 100μL。阴性对照用50%乙醇代替样品,空白对照用50%乙醇代替DPPH,其余操作均相同。充分混匀反应液,室温避光孵育30分钟。用酶标仪读取517nm吸光光度值,ΔOD值越低,表明样品对自由基的清除活性越高。计算DPPH自由基清除率和IC50。
1.2 ABTS自由基清除实验
本实验采用吸光光度法进行。在96孔板内加入样品,设3个复孔,每个样品2个稀释梯度,进行10倍倍比稀释(初筛后若样品具有明显的ABTS自由基清作用,则每个样品4个稀释梯度,进行2倍倍比稀释进行复筛),每孔100μL,每孔加入0.1M ABTS 100μL。阴性对照用50%乙醇代替样品,空白对照用50%乙醇代替ABTS,其余操作均相同。充分混匀反应液,室温避光孵育30分钟。用酶标仪读取734nm吸光光度值,ΔOD值越低,表明样品对自由基的清除活性越高。计算ABTS自由基清除率和IC50。
2.实验材料:
Trolox(Sigma,货号:238813-1G,批号:BCBP2767V)、DPPH(Sigma,货号:D9132-1G,批号:STBF5255V)、ABTS(Sigma,货号:A1888-1G,批号:SLBN7932V)、K2S2O8(Sigma,货号:P112194-25G,批号:F1508005)、磷酸二氢钠(西陇化工股份有限公司,批号:1202011)、磷酸氢二钠(西陇化工股份有限公司,批号:111021)、无水乙醇(利安隆博化(天津)医药有限公司货号:D4540-100ML,批号:20150205)、DMSO(最终反应体系中,DMSO的体积分数<1%)、Multiskan GO酶标仪(Thermo)、PRACTUM224-1CN电子天平(Satorius)、GNP-9270隔水式恒温培养箱(上海精宏实验设备有限公司)、30~300μL排枪(Thermo公司)、各量程移液枪(大龙公司)。
3.实验结果:
表1:DPPH自由基和ABTS自由基清除药效结果表
结果显示,1%DMSO(溶媒对照)对试验系统无明显影响;Trolox(阳性对照)的DPPH自由基清除IC50为6.97μg/mL,ABTS自由基清除IC50为7.77μg/mL;;黄芩、白梅花、丹参和白薇,单独使用均具有抗DPPH自由基和抗ABTS自由基功效;将4味药材混合精制形成组合物后抗DPPH自由基和抗ABTS自由基作用均明显增加,且制备成相关制剂仍有自由基清除作用。
Claims (8)
1.一种植物抗氧化组合物及其日化品,其特征在于,所述的植物抗氧化组合物包括如下重量份的组分:黄芩饮片1-40份、白梅花饮片1-40份、丹参饮片0.1-20份、白薇饮片0.1-20份。
2.根据权利要求1所述的一种植物抗氧化组合物及其日化品,其特征在于,该组合物的提取方法为:
分别取黄芩、白梅花、丹参饮片和白薇饮片,混合粉碎,加m:V比为3-20倍量的20-90%
乙醇回流提1-3次,每次30-180分钟,提取液合并,过滤,滤液浓缩,备用;
将上述浓缩液过大孔树脂柱,20-90%乙醇洗脱,洗脱液合并,干燥即得。
3.根据权利要求1-2所述的一种植物美白和抗氧化组合物及其日化品,其特征在于,该组合物用于日化品中的重量份为0.001-10份。
4.根据权利要求1-3所述的一种植物抗氧化组合物及其日化品,其特征在于,该组合物制成的日化品为面膜、精华液、护肤乳、护肤凝胶、洁面乳、沐浴乳,优选面膜、精华液、洁面乳。
5.根据权利要求1-4所述的一种植物抗氧化组合物及其日化品,其特征在于,含该组合物的面膜的制备方法为:
方法一:分别取重量份0.1-5份的植物抗氧化组合物、0.001-0.1份的虾青素,加入0.5-50份的二乙二醇单乙基醚、0.1-10份的聚山梨醇酯-80和0.05-5份的氢化卵磷脂混合均匀后,加入0.01-1份的水溶性神经酰胺、0.01-2份的卡波姆、0.01-2份的防腐剂、0.01-2份的香精和适量水,充分搅拌混合均匀,分装,即得;
方法二:分别取重量0.001-0.1份的植物抗氧化组合物、0.001-0.1份的虾青素、0.001-0.1份的辅酶Q10,加入0.5-50份的二乙二醇单乙基醚、0.1-10份的聚山梨醇酯-80和0.05-5份的氢化卵磷脂混合均匀后,加入0.01-10份的紫皮石斛提取物、0.01-10份的灵芝提取物、0.01-2份的黄原胶、0.01-5份的防腐剂、0.01-2份的香精和适量水,充分搅拌混合均匀,分装,即得;
方法三:分别取重量份0.1-10份的异构十二烷、0.1-5份的二异硬脂醇苹果酸酯、0.02-2份的聚二甲基硅氧烷、0.02-2份的聚甲基倍半硅氧烷、0.001-0.1份的虾青素、0.1-5份的乙二醇双硬脂酸酯、0.1-10份的乳木果油,加热混合均匀,加入0.1-5份的植物抗氧化组合物与0.5-5份的二乙二醇单乙基醚、0.2-2份的聚山梨醇酯-80的混合液,加热混合均匀,再加入0.001-0.1份的脯氨酸、0.001-0.1份的精氨酸、0.001-0.1份的缬氨酸、0.001-0.1份的苏氨酸、0.001-1份的透明质酸钠、0.5-5份的丙二醇、0.1-10份的单双硬脂酸甘油酯、0.1-10份的聚甘油-3双硬脂酸酯、0.1-10份的甘油聚乙二醇-75-硬脂酸酯、0.01-5份的防腐剂、0.01-2份的香精和适量水,保持60-100℃均质乳化5-60分钟,冷却,灌装,即得。
6.根据权利要求1-4所述的一种植物抗氧化组合物及其日化品,其特征在于,含该组合物的精华液的制备方法为:
方法一:分别取重量份0.1-10份的植物抗氧化组合物、0.001-0.1份的虾青素、0.001-0.1份的辅酶Q10,加入0.5-50份的二乙二醇单乙基醚、0.1-10份的聚山梨醇酯-80和0.05-5份的氢化卵磷脂混合均匀后,加入0.001-0.1份的脯氨酸、0.001-0.1份的精氨酸、0.001-0.1份的缬氨酸、0.001-0.1份的苏氨酸、0.02-0.2份的聚丙烯酰胺、0.01-2份的防腐剂、0.01-2份的香精和适量水,充分搅拌混合均匀,灌装,即得;
方法二:分别取重量0.1-10份的植物抗氧化组合物、0.001-0.1份的虾青素、0.001-0.1份的辅酶Q10,加入0.5-50份的二乙二醇单乙基醚、0.1-10份的聚山梨醇酯-80和0.05-5份的氢化卵磷脂、0.01-1份的乳木果油、0.01-5份的油酸聚乙二醇甘油酯混合均匀后,加入0.001-0.1份的脯氨酸、0.001-0.1份的精氨酸、0.001-0.1份的缬氨酸、0.001-0.1份的苏氨酸、0.01-10份的PEG-40氢化蓖麻油、0.01-2份的防腐剂、0.01-2份的香精和适量水,充分搅拌混合均匀,低温混合氮气,灌装,即得。
7.根据权利要求1-4所述的一种植物抗氧化组合物及其日化品,其特征在于,含该组合物的洁面乳的制备方法为:
分别取1-30份的肉豆蔻酸、月桂酸、硬脂酸和0.5-20份的甘油,加热融化,加入1-30份的氢氧化钾30-90℃搅拌5-180min,加入0.001-0.1份的植物抗氧化组合物、1-10份的丙二醇、0.1-10份的聚山梨醇酯-80加热混合,搅拌均匀后加入0.5-10份的椰油酰胺甜菜碱,0.5-10份的椰油酰两性基二乙酸二钠,0.5-10份的棕榈酸异丙酯,0.1-20份的二异硬脂醇苹果酸酯,0.5-10份的单硬脂酸甘油酯, 0.5-10份的鲸蜡硬脂基葡糖苷,0.01-3份的聚季铵盐-39,0.01-2份的防腐剂,0.01-3份的香精和适量水,保温混合,冷却,灌装,即得。
8.根据权利要求1-7所述的一种植物抗氧化组合物及其日化品,其特征在于,该植物组合物对常见的DPPH自由基和ABTS自由基均有不同程度的清除效果,具有对抗氧化损伤、延缓衰老、改善皮肤暗黄、减轻细纹、增加皮肤光泽度的功能,适用于全身各部位的清洁和维护。
