技术领域
本发明属于化学合成领域，具体为地拉罗司精制方法，更确切地说是4-[3,5-双(2-羟基苯基)-1H-1,2,4-三唑-1-基]-苯甲酸的精制方法。
背景技术
铁是人体必需的微量元素之一，其在体内含量的高低对维持细胞正常生长和代谢起着重要的作用，正常人每天铁需要量为20～25mg，人体全身的总铁量为3～4g，铁在体内分为功能铁和贮存铁，功能铁占全身总铁量的80%，贮存铁占铁总量约20%，血清铁蛋白是反映机体铁贮存情况的标志，是判断体内铁缺乏及铁超负荷的有效指标，人体铁元素超过一定的量就会造成危害，这种现象称为铁超负荷，通常认为SF>300μg/L、转铁蛋白饱和度>45%即可诊断为铁超负荷。据估计，目前全世界约有几百万人患有铁超负荷症。
铁超负荷是一种渐进性的、可以危及生命的疾病，常常是频繁输血的不可避免的并发症。某些慢性血液疾病需要长期、频繁的输血，这些患者很可能发生铁超负荷，如海洋性贫血、镰刀细胞贫血、骨髓异常增生综合征等。铁超负荷的征象可能在10～20次输血后就可显现。如果不加治疗，体内过量的铁会损害肝脏、心脏和内分泌腺。
地拉罗司，是首个1日一次的口服铁螯合剂，主要用于2岁及以上儿童和成人患者由输血导致的慢性铁超负荷。由瑞士诺华公司研制，2005年11月2日获得FDA批准上市，并于2005年12月12日在美国上市。在2010年6月获得SFDA批准进口，在2011年12月15号批准新适应症临床试验。
关于地拉罗司纯化和精制的工艺，已有相关文献报道，但多采用醇类溶剂析晶的方式，由于地拉罗司溶解性差，故这些方法存在溶剂量大、精制效果差等不足。
发明内容
本发明针对现有技术的不足，提供一种地拉罗司的精制方法，其具有精制效果好，收率高，操作简单，不可控制性低，经济实用的优点。
本发明的技术方案为：
地拉罗司的精制方法，其特征在于，包括以下步骤：
1）将地拉罗司粗品溶于四氢呋喃或其衍生物中，然后脱色，再加入有机溶剂，并洗涤有机相；
2）提取有机相，然后调节溶液pH为1～4，过滤；优选的，调节溶液pH为2～3；
3）将过滤所得固体打浆，过滤，回流打浆，冷却，过滤得地拉罗司精品。
优选的，所述四氢呋喃或其衍生物的与地拉罗司粗品的体积质量比为4～8:1，更优选6：1。
优选的，所述四氢呋喃或其衍生物为甲基四氢呋喃。
优选的，所述有机溶剂用量与四氢呋喃等体积。
优选的，所述有机溶剂选自异丙醚、二氯甲烷、正己烷、和/或石油醚。
优选的，所述脱色方法为活性炭脱色，保持回流0.5～2h，活性炭用量为地拉罗司质量的5%-15%。
所述洗涤的方法为：用pH小于10的弱碱性水溶液洗涤1～3次，其中弱碱选自：碳酸氢钠、碳酸钠、碳酸氢钾、碳酸钾等碱性无机盐水溶液。
优选的，所述洗涤方法为：用浓度为0.5%～2.0%碳酸氢钠水溶液洗涤1～3次。
优选的，所述提取选用的试剂为氢氧化钠水溶液，其用量为地拉罗司粗品质量的6～10倍，更优选8倍。
优选的，所述打浆的具体方法为：用20～40倍于拉罗司粗品质量的纯化水，在40℃条件下搅拌1～3h，过滤，重复操作1～4次。
优选的，所述回流打浆的方法为：用5倍于地拉罗司粗品质量的甲醇打浆，保持回流2h。
本发明有益效果在于：
本发明在步骤1）中采用活性炭进行脱色，脱色效果显著。弱碱性水溶液洗涤有机相，不仅有显著的脱色效果，还能有效除去地拉罗司主峰后杂质。
进一步地循环步骤1）和2），更有利于提高精制效果，达到更严格的质量标准。
进一步地采用水和甲醇打浆，可有效除去地拉罗司中的无机盐和地拉罗司主峰前的杂质。
本发明方法还具有适合工业化大批量生产，操作简单，质量可控，成本低等优点。

实施例一
地拉罗司精制工艺：
步骤1：向反应釜中加入300L四氢呋喃，再加入50Kg地拉罗司粗品（土黄色固体，HPLC：98.51%、16.54min杂质：0.054%、17.38min杂质：0.152%），搅拌至溶解，加入5Kg活性炭，加热至回流，搅拌1h，过滤，滤液冷却至室温后搅拌下加入300L异丙醚，再加入150L碳酸氢钠水溶液（1%，即1g碳酸氢钠溶于100ml水中），搅拌10min，静置分层，弃去水相，有机相再加入150L碳酸氢钠水溶液（1%），搅拌10min，精制分层，收集有机相。
步骤2：向上述有机相中加入400L氢氧化钠水溶液（1mol/L），搅拌20min，静置分层，收集水相，向水相中缓慢加入稀硫酸（2mol/L）约270L调pH=2～3，析出固体后继续搅拌1h，甩滤，滤饼干燥得45Kg类白色固体（HPLC：99.32%、16.54min杂质：0.006%、17.38min杂质：0.005%）。
步骤3：将上述固体加入到1800L纯化水中，40℃搅拌打浆1h，过滤，再用相同水量打浆两次。将所得固体加入到225L甲醇中，回流打浆2h，降温至室温，继续搅拌1h，甩滤，滤饼干燥得43Kg地拉罗司精品（HPLC：99.89%、16.54min杂质：0.006%、17.38min杂质：0.005%），收率86%。
实施例二
地拉罗司精制工艺：
步骤1：向反应釜中加入300L四氢呋喃，再加入50Kg地拉罗司粗品（土黄色固体，HPLC：99.10%、16.54min杂质：0.044%、17.38min杂质：0.264%），搅拌至溶解，加入5Kg活性炭，加热至回流，搅拌1h，过滤，滤液冷却至室温后搅拌下加入300L石油醚，再加入150L碳酸氢钠水溶液（0.5%，即1g碳酸氢钠溶于100ml水中），搅拌10min，静置分层，弃去水相，有机相再加入150L碳酸氢钠水溶液（0.5%），搅拌10min，精制分层，收集有机相。
步骤2：向上述有机相中加入400L氢氧化钠水溶液（1mol/L），搅拌20min，静置分层，收集水相，向水相中缓慢加入稀硫酸（2mol/L）约270L调pH=2～3，析出固体后继续搅拌1h，甩滤，滤饼干燥得48Kg（HPLC：99.31%、16.54min杂质：0.037%、17.38min杂质：0.100%）类白色固体。
重复进行步骤1，得有机相溶液。
重复进行步骤2，得类白色固体45Kg（HPLC：99.42%、16.54min杂质：0.012%、17.38min杂质：0.011%）。
步骤3：将上述固体加入到1720L纯化水中，40℃搅拌打浆1h，过滤，再用相同水量打浆两次。将所得固体加入到225L甲醇中，回流打浆2h，降温至室温，继续搅拌1h，甩滤，滤饼干燥得42Kg地拉罗司精品（HPLC：99.92%、16.54min杂质：0.011%、17.38min杂质：0.012%），收率84%。
实施例三
地拉罗司精制工艺：
步骤1：向反应釜中加入300L甲基四氢呋喃，再加入50Kg地拉罗司粗品（土黄色固体，HPLC：99.27%、16.54min杂质：0.025%、17.38min杂质：0.242%），搅拌至溶解，加入5Kg活性炭，加热至回流，搅拌1h，过滤，滤液冷却至室温后搅拌下加入300L异丙醚，再加入150L碳酸氢钠水溶液（1%，即1g碳酸氢钠溶于100ml水中），搅拌10min，静置分层，弃去水相，有机相再加入150L碳酸氢钠水溶液（1%），搅拌10min，精制分层，收集有机相。
步骤2：向上述有机相中加入400L氢氧化钠水溶液（1mol/L），搅拌20min，静置分层，收集水相，向水相中缓慢加入稀硫酸（2mol/L）约270L调pH=2～3，析出固体后继续搅拌1h，甩滤，滤饼干燥得47Kg类白色固体（HPLC：99.45%、16.54min杂质：0.015%、17.38min杂质：0.012%）。
步骤3：将上述固体加入到1800L纯化水中，40℃搅拌打浆1h，过滤，再用相同水量打浆两次。将所得固体加入到225L甲醇中，回流打浆2h，降温至室温，继续搅拌1h，甩滤，滤饼干燥得44Kg地拉罗司精品（HPLC：99.88%、16.54min杂质：0.014%、17.38min杂质：0.011%），收率88%。
实施例四
地拉罗司精制工艺：

步骤2：向上述有机相中加入400L氢氧化钠水溶液（1mol/L），搅拌20min，静置分层，收集水相，向水相中缓慢加入稀硫酸（2mol/L）约270L调pH=3～4，析出固体后继续搅拌1h，甩滤，滤饼干燥得46Kg类白色固体（HPLC：99.45%、16.54min杂质：0.012%、17.38min杂质：0.009%）。

步骤1：向反应釜中加入300L甲基四氢呋喃，再加入50Kg地拉罗司粗品（土黄色固体，HPLC：99.27%、16.54min杂质：0.049%、17.38min杂质：0.151%），搅拌至溶解，加入2.5Kg活性炭，加热至回流，搅拌1h，过滤，滤液冷却至室温后搅拌下加入300L二氯甲烷，再加入150L碳酸钠水溶液（0.5%，即0.5g碳酸钠溶于100ml水中），搅拌10min，静置分层，弃去水相，有机相再加入150L碳酸钠水溶液（0.5%），搅拌10min，精制分层，收集有机相。
步骤2：向上述有机相中加入400L氢氧化钠水溶液（1mol/L），搅拌20min，静置分层，收集水相，向水相中缓慢加入稀硫酸（2mol/L）约300L调pH=1～2，析出固体后继续搅拌1h，甩滤，滤饼干燥得45Kg类白色固体（HPLC：99.45%、16.54min杂质：0.012%、17.38min杂质：0.013%）。
步骤3：将上述固体加入到1350L纯化水中，40℃搅拌打浆1h，过滤，再用相同水量打浆两次。将所得固体加入到225L甲醇中，回流打浆2h，降温至室温，继续搅拌1h，甩滤，滤饼干燥得43Kg地拉罗司精品（HPLC：99.88%、16.54min杂质：0.011%、17.38min杂质：0.008%），收率86%。
对比例一
向反应釜中加入200L甲醇，再加入5Kg地拉罗司粗品（土黄色固体，HPLC：98.51%、16.54min杂质：0.054%、17.38min杂质：0.152%），加热至回流，搅拌至溶清，加入500g活性炭，搅拌1h，过滤，滤液冷却至5～10℃，搅拌析晶3h，甩滤，滤饼干燥得3.7Kg淡黄色固体（HPLC：98.66%、16.54min杂质：0.048%、17.38min杂质：0.145%），收率74%。
Claims (12)
1.一种地拉罗司的精制方法，其特征在于，包括以下步骤：
1)将地拉罗司粗品溶于四氢呋喃或甲基四氢呋喃中，然后脱色，再加入有机溶剂，并用弱碱性水溶液洗涤有机相；
2)提取有机相，然后调节提取相溶液pH为1～4，过滤；
3)将过滤所得固体打浆，过滤，回流打浆，冷却，过滤得地拉罗司精品，
其中所述有机溶剂选自异丙醚、二氯甲烷、正己烷和石油醚；提取有机相所用试剂为氢氧化钠水溶液。
2.根据权利要求1所述的地拉罗司的精制方法，其特征在于，调节提取相溶液pH为2～3。
3.根据权利要求1所述的地拉罗司的精制方法，其特征在于，所述四氢呋喃或甲基四氢呋喃与地拉罗司粗品的体积质量比L/Kg为4～8:1。
4.根据权利要求1所述的地拉罗司的精制方法，其特征在于，所述四氢呋喃或甲基四氢呋喃与地拉罗司粗品的体积质量比L/Kg为6：1。
5.根据权利要求1所述的地拉罗司的精制方法，其特征在于，所述有机溶剂用量与四氢呋喃等体积。
6.根据权利要求1所述的地拉罗司的精制方法，其特征在于，脱色方法为活性炭脱色，保持回流0.5～2h，活性炭用量为地拉罗司质量的5％-15％。
7.根据权利要求1所述的地拉罗司的精制方法，其特征在于，所述洗涤的方法为：用pH小于10的弱碱性水溶液洗涤1～3次，其中弱碱选自：碳酸氢钠、碳酸钠、碳酸氢钾和碳酸钾。
8.根据权利要求1所述的地拉罗司的精制方法，其特征在于，所述洗涤的方法为：用浓度为0.5％～2.0％的碳酸氢钠水溶液洗涤。
9.根据权利要求1所述的地拉罗司的精制方法，其特征在于，所述氢氧化钠水溶液的用量为地拉罗司粗品质量的6～10倍。
10.根据权利要求1所述的地拉罗司的精制方法，其特征在于，所述氢氧化钠水溶液的用量为地拉罗司粗品质量的8倍。
11.根据权利要求1所述的地拉罗司的精制方法，其特征在于，所述打浆的方法为：用20～40倍地拉罗司粗品质量的纯化水，在40℃条件下搅拌1～3h，过滤，重复操作1～4次。
12.根据权利要求1所述的地拉罗司的精制方法，其特征在于，所述回流打浆的方法为：用5倍于地拉罗司粗品质量的甲醇打浆，保持回流2h。
