The validity of the free-ion model (FIM) for the element lutetium (Lu), a member of the lanthanid... more The validity of the free-ion model (FIM) for the element lutetium (Lu), a member of the lanthanides, was assessed in experiments with the bacterium Vibrio fischeri. The FIM is mainly based on divalent metals and synthetic ligands and has not yet been validated for the trivalent lanthanides. The bioluminescence response of V. fischeri was studied at different Lu concentrations in the presence and absence of natural and synthetic organic ligands [citrate, malate, oxalate, acetate, ethylenediaminetetraacetate (EDTA), and nitrilotriacetate (NTA)]. All ligands were tested separately to ensure that their concentrations would not cause adverse effects themselves. Free Lu3+ concentrations were calculated with a speciation program, after extension of its database with the relevant Lu equilibria. The results confirmed the FIM for Lu: that is, in contrast to total dissolved Lu concentrations, free Lu3+ concentrations had an apparent relationship with the response of V. fischeri. However, a contribution of minor inorganic Lu complexes cannot be ruled out. In the presence of malate and oxalate, the EC50 for Lu3+ decreased faster in time than for the other ligands, indicating lower elimination rates. With an EC50 of 1.57 microM, Lu3+ is more toxic than La3+, Cd2+, or Zn2+ and approximately equally as toxic as Cu2+. Although the pH increased slightly during the experiments, it was shown that the influence of pH on Lu speciation was limited.

PCB 180 (2,2′,3,4,4′,5,5′-heptachlorobiphenyl) is a persistent and accumulating polychlorinated b... more PCB 180 (2,2′,3,4,4′,5,5′-heptachlorobiphenyl) is a persistent and accumulating polychlorinated biphenyl abundantly present in food and the environment. In this study, we used highly purified PCB 180 (dioxinlike impurities: 2.7 ng TEQWHO/g PCB 180) in a 28-day toxicity study in young adult Sprague-Dawley rats. Male and female rats were given total doses of 3, 10, 30, 100, 300, 1000 or 1700 mg/kg b.w. PCB 180 by gavage. Increased liver weights were observed at ≥300 mg/kg b.w. in males and females. No increases in serum ALT or ALP activities were found. A significant increase in liver pentoxyresorufin O-dealkylase (PROD) activity was found in males at ≥10 mg/kg b.w. and in females at ≥30 mg/kg b.w. In both genders, a significant induction of hepatic 7-ethoxyresorufin O-deethylase (EROD) activity was also observed in males at ≥10 mg/kg b.w. and in females at ≥300 mg/kg b.w. Western blotting showed that mainly cytochromes P450 (CYPs) 2B1/2 and 3A1 were induced while slight effects were seen on CYP1A1, CYP1A2 and CYP1B1. However, no induction of CYP1A1, 1A2 and 1B1 was found on the mRNA level, except for a slight effect in females at 1000 mg/kg b.w. Furthermore, hepatic UDP-glucuronosyltransferases (UGTs) 1A1 and 1A6 were markedly induced in males and slightly induced in females. The hepatic concentrations of apolar retinoids were decreased in males at ≥30 mg/kg b.w. and in females at ≥300 mg/kg b.w. Taken together our findings show that pure PCB 180 leads to hepatic changes in a dose range which did not cause CYP1A1 induction but causes centrilobular liver hypertrophy, affects drug-metabolizing enzymes involved in the metabolism of exogenous and endogenous substrates and leads to changes in liver retinoid levels. A benchmark dose (BMD) approach is presented in order to model lowest effective dose levels for these effects. Comparison of PCB 180 liver level related to BMDL5 for hepatic hypertrophy in rats with human data on ‘total′ hepatic PCB levels in individuals without history of specific exposure suggests a relatively small margin of tissue burden in the range of 37-fold. In this study we developed a new LC nanofractionation platform that combines a human cell (BG1.Lu... more In this study we developed a new LC nanofractionation platform that combines a human cell (BG1.Luc) gene reporter assay with a high resolution mass spectrometer for the detection and identification of estrogenic and anti-estrogenic compounds in environmental waters. The selection of this assay was based on its high sensitivity and selectivity, which is required for environmental trace level detection. We modified an autosampler and controlled it with in-house developed software to collect fractions in the low second range in microtiter plates. This ensured that chromatographic separation was maintained and allowed straightforward hyphenation with the bioassay. After bioassay testing, bioassay chromatograms were reconstructed and directly correlated with MS chromatograms that were obtained in parallel. This enabled to pinpoint bioactives in the MS chromatogram within a single fractionation cycle and results in a significant increase in throughput compared to traditional EDA studies. The sensitivity of the platform was low enough for environmental waters (80nM for bisphenol A and 320pM and 3.2nM for estradiol and estriol, respectively). In addition, the ability of the platform to detect anti-estrogens was successfully demonstrated as well. Finally, real samples were analysed.

The most widely described endpoint is estrogenicity, but other hormonal disturbances, e.g., thyroid hormone disruption, are gaining more and more attention. Here, we present a review and chemical characterization, using principal component analysis, of organic compounds that have been tested for their capacity to bind competitively to the thyroid hormone transport protein transthyretin (TTR). The database contains 250 individual compounds and technical mixtures, of which 144 compounds are defined as TTR binders. Almost one third of these compounds (n = 52) were even more potent than the natural hormone thyroxine (T4).
