Corn plantation produces waste in the form of corncob. Corncob is lignocellulosic biomass that can converted to cellulose, even sugar or another chemical like ethanol. The first step for utilization of corncob is delignification. The purpose of this study was to isolate and purify the enzyme laccase from white rot fungus Cerrena sp.B.Md.T.A.1 that inoculation in GDP media used for biodelignification of corncobs. Enzymatic biodelignification conduct with the variation of substrate was 5%, 10%, and 15% (w/v), while the time variation were 6 hours and 24 hours. The percentage of lignin concentration before pretreatment reached is 37.25%. The highest reduction of lignin content was achieved in 15% substrate for 24 hours incubation time around 42.71%.

Objective: To fractionate and identify polyphenols from Guazuma ulmifolia Lam. leaves, and to exp... more Objective: To fractionate and identify polyphenols from Guazuma ulmifolia Lam. leaves, and to explore their antioxidant, 5-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase inhibitory, and Nrf2 modulatory activities. Methods: The 1,1-diphenyl-2-picrylhydrazyl assay was used to evaluate the antioxidant activity of a polyphenolic fraction of the extract of Guazuma ulmifolia Lam. leaves. THP-1 gene reporter cell lines constructed with a transcriptional response element specific for Nrf2 and a minimal promoter for the firefly luciferase–green fluorescent protein transgene were used to determine the effect of the polyphenolic fraction on the Nrf2 signaling pathway. Furthermore, an assay of HMG-CoA reductase inhibitory activity was performed by using a commercial enzyme kit. Polyphenolic compounds were identified by liquid chromatography-tandem mass spectrometry. Results: The polyphenolic fraction showed fairly strong antioxidant activity [IC50 = (14.90 ± 4.70) μg/mL] and inhibited HMG-CoA reductase activity by 69.10%, which was slightly lower than that by pravastatin (84.37%) and quercetin (84.25%). Additionally, the polyphenolic fraction activated the Nrf2 antioxidant signaling pathway at 500 μg/mL. Eleven subfractions resulting from the column chromatography separation of the polyphenolic fraction also showed relatively strong antioxidant activities (IC50: 17.46μ217.14 μg/mL). The subfraction (F6) stimulated the Nrf2 signaling pathway and had HMG-CoA reductase inhibitory activity (65.43%). Moreover, the subfraction contained two main flavonoids: quercetin and quercimeritrin. Conclusions: The polyphenolic fraction of Guazuma ulmifolia could induce antioxidant genes via the Nrf2/antioxidant regulatory elements pathway, and is a promising candidate for an inhibitor of HMG-CoA reductase.

This study was conducted to investigate the decolorization of dyes using laccase produced by Cerrena sp. BMD.TA.1 isolated from Gunung Rinjani National Park (GRNP), West Nusa Tenggara – Indonesia. This fungus had capability to decolorize three types of dyes, those: anthraquinone (Remazol Brilliant Blue R, RBBR), azo (Congo Red) and triphenylmethane (Fast Green FCF). The optimum laccase activity in this dyes treatment by fungal culture was achieved at 96 h pre-incubation, but the highest decolorization rate was reached at 144 h pre-incubation. After 72 h of dyes treatment, all dyes were removed at least 82%. The optimum decolorization for 100 mg L−1 was similar for of all dyes types. The aims of this research were to analyze secondary metabolite compounds and determine total phenol and total flavonoid of Melastoma affine fruit extract. The samples used in this study were unripe and ripe Melastoma affine fruits. Both of samples were extracted by three different solvents (water, 70% ethanol, and 96% ethanol) and obtained 6 extracts. Based on phytochemical test, each extract from unripe and ripe Melastoma affine fruit contained alkaloid, triterpenoid, flavonoid, phenol, and tanin. Penelitian ini bertujuan menganalisis komponen-komponen fitokimia daun suren secara kualitatif dan kuantitatif, serta menguji aktivitas sitotoksisitasnya terhadap sel vero dan sel kanker payudara MCF-7. Pelarut yang digunakan untuk mengekstraksi daun suren adalah air, etanol 70%, etil asetat, dan n-heksana. Rendemen yang diperoleh untuk pelarut air, etanol 70%, etil asetat, dan n-heksana secara berurutan sebesar 33,54; 34,85; 6,38; dan 1,31%. Komponen fitokimia dalam daun suren meliputi alkaloid, triterpenoid, flavonoid, tanin, fenol, dan steroid. Red yeast rice has many biological activities such as antihyperglycemic, antihypercolesterolemic,... more Red yeast rice has many biological activities such as antihyperglycemic, antihypercolesterolemic, antidiabetic and antioxidant. This study was conducted to search for the most active extracs of red yeast rice as an in vitro antidiabetes. Antidiabetic activity was measured through α-glucosidase inhibition assay. Samples were ethanol 95% extract of red yeast rice, as well as its fractionated products using n-hexane, dicloromethane, ethyl acetate and water. Extraction of red yeast rice powder with 95% ethanol had a yield of 9.44%. The yield from fractionation of ethanol 95% extract using n-hexane, dicloromethane, ethyl acetate and the water were 1.99%, 1.13%, 0.59% and 1.36% respectively. The phytochemical test showed that ethanol 95% extract of extract contained flavonoids, alkaloids and saponin.
