Applying MALDI-TOF MS, HPTLC and ESI-IT MS, we comparatively analyzed the PL compositions of semen and blood of species differing in their reproductive systems and types of nutrition (bull, boar, stallion, lion and man) with regard to the sensitivity to oxidation as well as the accumulation of harmful lyso-PL (LPL), transient products of lipid oxidation. In addition, the protective capacity of seminal fluid (SF) was also examined. The PL composition of erythrocytes and blood plasma is similar across the species, while pronounced differences exist for sperm and SF. Since the blood function is largely conserved across mammalian species, but the reproductive systems may vary in many aspects, the obtained results suggest that the PL composition is not determined by the type of nutrition, but by the relatedness of species and by functional requirements of cell...

In the present study, heart rate (HR), respiratory rate (RR) and body temperature (BT) were evaluated in healthy mares of different breed types and compared to reference values for adult horses from the literature. During daily physical exams of periparturient mares, the HR, RR and BT were evaluated. Differences according to the horse's size were investigated and in large breeds, the influence of dystocia or retained placenta was analysed. During the last weeks before parturition (a. p.), the HR significantly increased and was clearly lower after parturition (p. p.; p < 0.05). In larger horses, the RR increased a. p. and decreased p. p. (p < 0.05). The BT underwent changes in all groups during the periparturient period and was higher p. p. (p < 0.05). Die Tiefgefrierkonservierung von Nebenhodenspermien stellt eine gute Möglichkeit zur Erhaltung de... more Die Tiefgefrierkonservierung von Nebenhodenspermien stellt eine gute Möglichkeit zur Erhaltung der Genreserve wertvoller Vatertiere dar, wenn ein plötzlicher Unfall eine herkömmliche Samengewinnung unmöglich macht. Im vorliegenden Fall wird von der Geburt eines gesunden Fohlens berichtet, das aus der Besamung mit Nebenhodenspermien entstand. Die Hoden und Nebenhoden wurden in Allgemeinanästhesie entfernt und gekühlt über Nacht in das Labor verbracht. Durch retrograde Spülung der Nebenhodenschwänze konnten 17,79 × 109 Spermien konserviert werden. Eine fruchtbare Stute wurde 1 Jahr später mit tiefgefrorenen Nebenhodenspermien tief intrauterin besamt. Im 1. Zyklus wurde die Stute nicht tragend. Im 2. Zyklus wurde homologes Seminalplasma zusammen mit den Nebenhodenspermien in den Uterus verbracht und 14 Tage nach der Ovulation konnte eine Trächtigkeit festgestellt werden. In der Studie wurde untersucht, ob sich bei gesunden Stuten unterschiedlichen Pferdetyps in der Spätträchtigkeit und in der 1. Woche post partum (p. p.) die Herz- (HF) und Atemfrequenz (AF) sowie die innere Körpertemperatur (IKT) von den in der Literatur angegebenen Referenzwerten für adulte Pferde unterscheiden. Material und Methoden: Im Rahmen der täglichen klinischen Untersuchung erfolgte eine Auswertung der HF, AF und IKT von Stuten, die zur Geburtsüberwachung stationär eingestellt waren. Unterschiede in Bezug auf die Pferdegröße wurden ausgewertet und bei Großpferden wurde der Einfluss einer Schwergeburt und von Nachgeburtsverhalten evaluiert. Clinical, histological, and postmortem examination revealed an underdeveloped genital tract, the absence of a cervix uteri, and small inactive ovaries without male gonadal tissue. Blood lymphocyte analysis revealed an unusual mosaic karyotype consisting of 2 cell lines. For the majority of cells (70%), monosomy X (63,X) was observed. The remaining cells (30%) contained 64 chromosomes including one X chromosome and a small rudimentary Y chromosome consisting mostly of heterochromatin. The centromere was retained, but its full functionality was questionable. PCR analysis revealed that the entire male-specific region of Y (Yq14), including the SRY gene, was deleted. It remained unclear if the pseudoautosomal region (Yq15) and parts of the heterochromatic region (Yq13) were affected by this deletion. Abstract Insemination with chilled transported semen has become distinctly important in the horse... more Abstract Insemination with chilled transported semen has become distinctly important in the horse‐breeding industry. To ensure cell survival during cooled storage, semen is diluted with an appropriate extender and the concentration of seminal plasma (SP) is reduced. Nevertheless, SP plays an important immunomodulatory role in the female genital tract and supports sperm fertility. The aim of the present study was to evaluate the effect of the addition of autologous SP after cooled storage to highly concentrated stallion semen. Therefore, SP was removed by simple centrifugation of extended semen, aspiration of the supernatant, and resuspension of the sperm pellet with semen extender. Motion characteristics were evaluated after cooled storage for 48 hours at concentrations of 333 × 106 sperm/mL in comparison with stored samples at concentration of 25 × 106 sperm/mL (control). The highly concentrated semen samples were diluted with an extender containing 0%, 5%, 20%, and 80% SP directly before motility analysis. Dilution of the cooled semen with a fresh semen extender without SP (0%) increased kinematic parameters (curvilinear velocity [VCL] 137.3 vs. 151.8; straight‐line velocity [VSL] 49.0 vs. 57.5; average path velocity [VAP] 69.5 vs. 79.4 &mgr;m/second; amplitude of lateral head [ALH] 3.1 vs. 3.3 &mgr;m; beat cross frequency [BCF] 31.6 vs. 33.5 Hz; P < .05) but not total motility (51% vs. 43%) and progressive motility (46% vs. 36%) compared with controls. The addition of SP after storage for 48 hours decreased sperm total motility and progressive motility regardless of SP concentration: 5 (38% and 34%), 20 (37% and 33%), and 80% SP (27% and 22%; P < .05). In contrast, kinematic parameters were enhanced by extenders containing 5% and 20% SP (VCL: 148.0 and 155.6; VSL: 59.2 and 60.9; VAP: 78.7 and 81.9; BCF: 33.4 and 35.7; ALH: 3.4 and 3.4; P < .05). However, using an extender containing 80% SP was detrimental to kinematic parameters (VCL: 151.2; VSL: 52.2; VAP: 76.9; BCF: 34.8; P < .05) except for ALH, which increased (3.5; P < .05). In conclusion, cooled storage at concentrations of 333 × 106 sperm/mL did not affect sperm motility. The addition of a fresh extender or an extender containing small concentrations of SP to highly concentrated ejaculated sperm increased kinematic values after storage; however, increasing concentrations of SP decreased sperm motility. Necropsy revealed melanomas next to the brain stem but without evidence of primary dermal melanoma at most common sites as perineal region, tail or external genitalia. Histological examination of tumorous masses revealed melanocytes with cellular and nuclear pleomorphism. Melanocytic infiltrations could also be detected in the musculus parotidoauricularis, in the plexus chorioideus, in the leptomeninges and in walls of celebral vessels. Immunohistochemical staining of HMB-45 revealed specific labelling of tumour cells and reverse transcriptase PCR showed expression of both, gp75 and gp100 in the tumour tissue.
