A germinacao pode ser util para conversao de isoflavonas ?-glicosidicas em agliconas. O objetivo deste trabalho foi investigar as alteracoes no perfil de isoflavonas no processo de germinacao da cultivar de soja BRS 284. A soja foi germinada por 168 h em rolos de papel de germinacao a 35 °C e 100 % de umidade relativa com luz natural. A cada 24 h as diferentes formas de isoflavonas foram quantificadas por CLUE. O experimento da germinacao foi conduzido em blocos ao acaso e os dados analisados por ANOVA e teste de Tukey (p<0,05). A soja nao germinada apresentou o perfil de isoflavonas em 100 g de amostra: 430,36 mg de isoflavonas totais; 97,66 mg de glicosidicas; 329,88 mg de malonilglicosideos e 2,82 mg de agliconas. However, isoflavones as aglycone have been regarded as the most bioactive form. In the present study, the profile of isoflavones and their scavenging activity as affected by germination were investigated in several soybean fractions, namely cotyledons, epicotyls, radicles, and hypocotyls. Only aglycones were detected in the radicles from 144 h until 168 h of germination, which makes this component a potential feedstock for studies aiming at isolation, especially of daidzein, which was present in higher concentrations. In terms of total yield and contribution to the total weight of the germinated soybeans, the cotyledons are the best source of aglycones, which was achieved at 144 h of germination. The aim of this study was to investigate the b-glucosidase activity of radicles, hypocotyls, coty... more The aim of this study was to investigate the b-glucosidase activity of radicles, hypocotyls, cotyledons and epicotyls of soybeans (Glycine max) grown in the presence and absence of light and to obtain an alternative source of b-glucosidase. Seeds were germinated in the presence and absence of light at various time points, and the radicles, epicotyls, hypocotyls and cotyledons were separated. The enzymatic activity in the epicotyls presented the highest activity at 144 h in the presence of light. Specifically, the enzymatic activity of the epicotyls was sixfold higher than that of radicles and 12-fold higher than that of cotyledons. Interestingly, the specific activity of hypocotyls and cotyledons was 4.02-fold and 5.17-fold lower than that of epicotyls after 144 h of germination under light. The high specific activity of lightgerminated epicotyls suggested that the purity of b-glucosidase from epicotyls is greater than b-glucosidase obtained from other parts of the plant. Isoflavones are beneficial to human health. Specifically, isoflavones and estrogen have similar positive effects on the symptoms of menopause and osteoporosis. Moreover, isoflavones possess antioxidant and antitumoral properties against certain cancers. In nature, the most common form of the isoflavone is the glycosylated form. Studies on the bioavailability of isoflavones indicate that these compounds behave similarly to estrogen in most biological systems and the aglycone forms are more bioavailable than their glycoside forms. b-glucosidase is produced from the metabolism of fungi, bacteria or soybeans, and is the only enzyme capable of hydrolyzing isoflavone glycosides to aglycones. Therefore, safe and effective methods for the synthesis of b-glucosidase must be developed to produce soy-based foods with high concentrations of aglycone isoflavones. Thus, the objective of the present study is to investigate the best conditions under which the greatest amount of b-glucosidase could be isolated from soybean.

The objective of this study was to optimize the extraction of different isoflavone forms (glycosi... more The objective of this study was to optimize the extraction of different isoflavone forms (glycosidic, malonyl-glycosidic, aglycone and total) from defatted cotyledon soy flour using the simplex-centroid experimental design with four solvents of varying polarity (water, acetone, ethanol and acetonitrile). The obtained extracts were then analysed by high-performance liquid chromatography. The profile of the different soy isoflavones forms varied with different extractions solvents. Varying the solvent or mixture used, the extraction of different isoflavones was optimized using the centroid-simplex mixture design. The special cubic model best fitted to the four solvents and its combination for soy isoflavones extraction. For glycosidic isoflavones extraction, the polar ternary mixture (water, acetone and acetonitrile) achieved the best extraction; malonyl-glycosidic forms were better extracted with mixtures of water, acetone and ethanol. Aglycone isoflavones, water and acetone mixture were best extracted and total isoflavones, the best solvents were ternary mixture of water, acetone and ethanol.

Furthermore, this enzyme may improve the conversion of isoflavones into their corresponding aglycones. β-Glucosidase may also increase the release of aglycones from the cell wall of the plant materials. Therefore, the aim of this work was to optimize both the extraction of β-glucosidase from EGS and to further examine its application in defatted soybean cotyledon to improve the recovery of aglycones, which were evaluated by ultra-high performance liquid chromatography (UHPLC). A multistep optimization was carried out and the effects of temperature and pH were investigated by applying a central composite design. The linear effect of pH and the quadratic effect of pH and temperature were significant for the extraction of β-glucosidase and recovery aglycones, respectively.
