S evidence of probiotic benefits on human health is continuously expending but many of the trends... more S evidence of probiotic benefits on human health is continuously expending but many of the trends currently affecting the probiotics market are related to the global regulatory movement. Although hundreds of products are marketed as probiotics, many of the strains they contain haven’t been evaluated sufficiently. In addition, there are still no official or generally accepted methods for the analysis of probiotic products, where quantification and strain identification are crucial. Legislation on food supplements is partially harmonized at the EU level in the frame of a regulation on nutrition and health claims made on foods. Perhaps the most difficult challenge for research on probiotic products is providing proof of efficacy in healthy individuals. In clinical trials, the efficiency of probiotic intervention has to be relied on well-documented effects measured with reliable biomarkers independent of the subjective criteria according to which a subject feels healthy. Where pathogenesis is not present or it is not completely understood, the efficacy of probiotic treatment is often evaluated according to the perceived improvement of quality of life. A wide variety of host, dietary and environmental factors that affect gastrointestinal micro biota additionally make clinical studies difficult. The significant effect of probiotic prevention or supportive treatment failed frequently due to small number of participants in the survey. As EFSA rejected all general claims about probiotics so far, food supplements are at a crossroads. Many recent probiotic studies are already focused to specific health problems instead of general well-being and it might be expected that increasing number of probiotic products will be registered as drugs. An online survey was conducted to gather data on the knowledge of health professionals. The online survey was distributed via email and social media platforms using snowball sampling. A total of 1066 health professionals (859; 80.6% female) from 30 countries responded to the survey. Most of the respondents evaluated their knowledge of probiotics as medium (36.4%) or good (36.2%). Only 8.9% of the respondents rated it as excellent. No statistical difference in knowledge was found between male and female health professionals. Over 80% of pharmacists, allied health professionals, medical doctors and dentists, and other health professionals knew the correct definition of probiotics as “live microorganisms that, when administered in adequate amounts, confer a health benefit on the host”, whereas three quarters of registered nurses and midwives and less than two thirds of psycholog...

The current study aims to clarify the signalling pathways and evaluate the possible beneficial effects of the combination of two strains. We used a dextran sulphate sodium (DSS)-induced mouse model of colitis. RNA extracted from the middle part of the colon tissue was used for examination of the global gene expression with Affymetrix microarrays. An enrichment analysis of the KEGG pathways was performed, and a subset of genes associated with intestinal epithelial barrier function was verified with qPCR. A clinical condition assessment of the differently treated mice revealed that the combination of these two bacterial strains was safe for use as a dietary supplement. Feces, collected at 3, 30, and 90 days after birth from 60 breastfed Slovenian newborns, was submitted to microbial DNA extraction and qPCR quantification of selected gut associated taxa. Multivariate regression analysis was applied to evaluate microbial dynamics with respect to infant demographic, environmental, clinical characteristics and first year growth data. Early microbial variability was marked by the proportion of Bacilli, but diminished and converged in later samples, as bifidobacteria started to prevail. The first month proportions of enterococci were associated with maternity hospital locality and supplementation of breastfeeding with formulae, while Enterococcus faecalis proportion reflected the mode of delivery. Culture-dependent analysis, followed by sequencing of 16S ribosomal DNA for bacteria and 26S rRNA gene for yeasts, revealed 3 different species of lactobacilli and yeasts, respectively. The most frequently isolated bacterial species were Lactobacillus kefiranofaciens ssp. kefirgranum, Lb. parakefiri, and Lb. kefiri, whereas yeasts belonged to Kluyveromyces marxianus, Kazachstania exigua, and Rhodosporidium kratochvilovae. This study is the first to report on the presence of R. kratochvilovae in kefir grains.
