There are profound species differences in IGF system expression and actions in reproduction. Use of multiple models and approaches is required to elucidate fundamental aspects of IGFs in nutrition and reproduction of humans and domestic animals.

P450 aromatase (P450arom, CYP19), a CYP19 gene product, is a member of the cytochrome P450 superf... more P450 aromatase (P450arom, CYP19), a CYP19 gene product, is a member of the cytochrome P450 superfamily that catalyzes the formation of aromatic C(18) estrogen from C(19) androgen. To begin to understand the molecular mechanisms of P450 aromatase action in the protogynous wrasse, we isolated two cDNAs: one encoding CYP19a from ovary and the other encoding CYP19b from brain. The full-length cDNA of wrasse CYP19a, isolated from ovary cDNA library, is 2020 bp long and encodes 519 amino acids. The amino acid sequence of CYP19a has 62-83% identity with ovary-type aromatases of other teleosts. The full-length cDNA of wrasse CYP19b obtained using 5' and 3' RACE consists of 2666 bp, and its open reading frame encodes 496 amino acids. The deduced amino acid sequence has 62-83% identity with brain-type aromatases of other teleosts. Northern blot analysis identified a single 2.2-kb transcript in the ovary (CYP19a), and a single 2.6-kb transcript in the brain (CYP19b), suggesting that there are single forms of CYP19a and CYP19b, respectively, in the wrasse. RT-PCR assay showed that two CYP19 genes were expressed ubiquitously in various tissues, although each CYP19 subtype was expressed at highest level in the ovary and brain of the wrasse. These results suggest that CYP19 genes act in diverse tissue types, in addition to their effects on the physiological and reproductive functions of estrogen.

This study evaluated the dietary effects of SM on serum cholesterol-related and serum antioxidant indexes in rats of both sexes. Sprague-Dawley rats [38 dams and their offspring (20 males and 20 females/diet)] were fed diets containing 0 (control), 1%, 4%, or 10% (wt:wt) SM powder from gestation day 4 through to postnatal day (PND) 126. Biochemical indexes were monitored during the midgrowth phase (PNDs 50-66). The food consumption by offspring fed the control diet and diets supplemented with SM was not different when measured on PND 65. The mammary tumor-protective effects of dietary factors are considered to be mediated by multiple... more The mammary tumor-protective effects of dietary factors are considered to be mediated by multiple signaling pathways, consistent with the heterogeneous nature of the disease and the distinct genetic profiles of tumors arising from diverse mammary cell populations. In a 7,12-dimethylbenz(a)anthracene-induced model of carcinogenesis, we showed previously that female Sprague-Dawley rats exposed to AIN-93G diet containing whey protein hydrolysate (WPH) beginning at gestation Day 4 had reduced tumor incidence than those exposed to diet containing casein (CAS), due partly to increased mammary differentiation and reduced activity of phase I metabolic enzymes. Here, we evaluated the tumor-protective effects of these same dietary proteins to the direct-acting carcinogen N-methyl-N-nitrosourea (NMU). We found that lifetime exposure to WPH, relative to CAS, decreased mammary tumor incidence and prolonged the appearance of tumors in NMU-treated female rats, with no corresponding effects on tumor multiplicity. At 115 days post-NMU, histologically normal mammary glands from WPH-fed tumor-bearing rats had increased gene expression for the tumor suppressor BRCA1 and the differentiation marker kappa-casein than those of CAS-fed tumor-bearing rats. Tumor-bearing rats from the WPH group had more advanced tumors, with a greater incidence of invasive ductal carcinoma than ductal carcinoma in situ and higher serum C-peptide levels than corresponding rats fed CAS. WPH-fed tumor-bearing rats were also heavier after NMU administration than CAS tumor-bearing rats, although no correlation was noted between body weight and C-peptide levels for either diet group. Results demonstrate the context-dependent tumor-protective and tumor-promoting effects of WPH; provide support for distinct signaling pathways underlying dietary effects on development of mammary carcinoma; and raise provocative questions on the role of diet in altering the prognosis of existing breast tumors.
