Les organismes photosynthetiques possedent de remarquables mecanismes de photoregulation et d'... more Les organismes photosynthetiques possedent de remarquables mecanismes de photoregulation et d'adaptation leur permettant d'optimiser leur machinerie photosynthetique. Ce travail a contribue a la comprehension des adaptations membranaires et moleculaires de cette machinerie chez des cyanobacteries, en fonction de l'intensite et de la qualite de la lumiere. L'utilisation du mutant desa /desd de synechocystis 6803, deficient en acides gras polyinsatures qui a pour consequence une rigidite membranaire a des temperatures plus elevees que chez le sauvage, nous a permis de conclure que la fluidite membranaire joue un role important dans le mecanisme des transitions d'etat. Nous avons aussi mis en evidence un quenching de fluorescence du psii induit par des fortes intensites de lumiere bleue lie a un mecanisme different de celui observe par la photoinhibition et par les phenomenes des transitions d'etat. Nous avons etudie ensuite le role de la lumiere comme agent regulateur de l'expression du gene psba (gene du psii) et du genepsae (gene du psi). En utilisant le mutant pal ( apcab, pc, apce) deficient des phycobilisomes, nous avons montre que l'etat redox de certains transporteurs d'electrons joue un role important dans la regulation de l'expression et que les photorecepteurs ne sont pas impliques dans cette regulation. Nous proposons que la qualite de la lumiere regule l'expression des genes via un controle redox du complexe cyt b6f.
CD4+ T cells have been found to play critical roles in the control of both acute and chronic Toxo... more CD4+ T cells have been found to play critical roles in the control of both acute and chronic Toxoplasma infection. Previous studies identified a protective role for the Toxoplasma CD4+ T cell-eliciting peptide AS15 (AVEIHRPVPGTAPPS) in C57BL/6J mice. Herein, we found that immunizing mice with AS15 combined with GLA-SE, a TLR-4 agonist in emulsion adjuvant, can be either helpful in protecting male and female mice at early stages against Type I and Type II Toxoplasma parasites or harmful (lethal with intestinal, hepatic, and spleen pathology associated with a storm of IL6). Introducing the universal CD4+ T cell epitope PADRE abrogates the harmful phenotype of AS15. Our findings demonstrate quantitative and qualitative features of an effective Toxoplasma-specific CD4+ T cell response that should be considered in testing next-generation vaccines against toxoplasmosis.
