Infectious bronchitis virus (IBV) has a significant impact on the poultry industry worldwide and ... more Infectious bronchitis virus (IBV) has a significant impact on the poultry industry worldwide and affecting all chicken ages, types and breeds causing severe economic losses. Genetic characterization showed that the recent Egyptian IBV strains continually evolved over time and at least two groups of viruses are circulating (Classic Variant II) which are distinctly apart from each other .Antigenic results revealed that there is cross protection between variant II strains and it needs more study to evaluate the real protection using challenge test and there is a very weak cross protection between the classic and variant II strains according to Cross HI results.Furthermore variable pathogenicity indexes were recoded among the tested IBV viruses

Peste des petits ruminants (PPR) is an economically important, highly contagious viral disease of... more Peste des petits ruminants (PPR) is an economically important, highly contagious viral disease of sheep and goats characterized by high morbidity and mortality rates. A live attenuated PPR vaccine has been produced in Veterinary Serum and Vaccines Research Institute (VSVRI), Abbasia, Cairo, Egypt. It has and still been utilized to control PPR disease in endemic areas of the Arabian Gulf. In this study, the identity of four batches of PPR vaccines was tested as per the Office International des Epizooties (OIE) guidelines using RT-PCR technique that based on the amplification of fragments of F-protein and fragments of N-protein. All four batches are also tested for identity using real time RT-PCR. Results in this study showed that molecular techniques could be used for rapid evaluation of PPR vaccine including RT-PCR and real time RT-PCR for identity testing. Equid alphaherpesvirus 1 (EHV-1) is the most important virus causing pathological disorders in ho... more Equid alphaherpesvirus 1 (EHV-1) is the most important virus causing pathological disorders in horses and causes persistent outbreaks of upper respiratory tract infection, ocular affections, abortion, and neurological disorders with high mortality in Arabian horses in Egypt. As EHV-1 is a very contagious disease, the quick and accurate diagnosis are important to broaden our understanding of EHV-1 in the field, implicate strong preventive, and control measures. Sixty-six samples collected over a period of 4 years were examined. Most of samples originated from Cairo and Giza governorates from respiratory, abortigenic and neurological outbreaks. EHV-1 was diagnosed in these clinical cases by immunohistochemistry using monoclonal antibody against EHV-1 glycoprotein B and molecular detection using gB, ORF33 specific real-time PCR. Molecular characterization of glycoprotein B (gB, ORF33) gene was applied for confirmation. Molecular characterization revealed that the ORF33 sequences of this study were identical. These sequences were closely related to the European EHV-1 strains. Furthermore, EHV-1 sequences in this study showed little to no differences for the amino acid sequences compared to previously published sequences. This study would be valuable for monitoring of EHV-1 infection in Egypt and determining the gB gene sequence of newly identified EHV-1 field strains which is the most conserved region in the viral DNA and frequently used as a target for diagnostic PCR protocols for the future outbreaks.

Methods: Blood sera, tracheal swabs and tissue organs were collected from 60 poultry flocks from different ages, different type of raising which were housed in different Governorates in Lebanon including, broilers, layer hens of breeders, backyard chickens and ducks for serosurveillance and molecular characterization. Cattle egret is one of the free-living birds that is often present near commercial chicken farms with a possibility of catching avian viral infections specially H5N1 AIV; this is augmented by broken biosecurity which is often in small and medium sized poultry production facilities. This study investigates the possible isolation of H5N1 AIV from cattle egrets in the spread and epidemiology and to try virus isolation if possible. Methods: rapid test strip for detecting avian influenza antigen was used to test 15 tracheal pool samples collected from 15 commercial broiler flocks showing rapid increase in mortality, respiratory and enteric manifestation in Desouk and Qallin, Kafr-elshiekh Province, Egypt from 2014 to 2015. The fulllength DNA copies of RNA segment 6 (coding for VP6 protein) of NCDV were inserted into a baculovirus expression vector. A recombinant baculovirus carrying the VP6 gene was constructed through homologous recombination between the baculovirus recombinant plasmid carrying the VP6 gene and Autographa californica nuclear polyhedrosis virus (AcNPV) under the control of the polyhedrin promotor. Infection of Spodoptera frugiperda (Sf9) cells with the recombinant baculovirus expressing VP6 protein revealed a high-level of expression when tested by immunoflurescence and solid phase ELISA tests using BRV-specific polyclonal antibodies.
